摘要 目的评价Epi Derm体外皮肤刺激性试验方法的可行性。方法取Epi Derm皮肤组织,加30μL受试样品,每个样品平行操作3个,同时设阴性对照和阳性对照,作用60 min,然后进行MTT染色、提取、570 nm测定吸光度值,计算皮肤组织细胞存活率,进而判定受试物的刺激性。同时与体内家兔急性皮肤刺激性试验结果进行比较。结果与体内家兔急性皮肤刺激性试验结果比较,Epi Derm体外皮肤刺激性试验方法灵敏度、特异度、准确度均达到100%,且该方法测量结果客观、工作量小、避免了动物伦理问题。结论 Epi Derm体外皮肤刺激性试验方法是目前最理想的体外皮肤刺激性替代方法。 Objective To assess the sensitivity, specificity and accuracy of an in vitro EpiDerm Skin Irritation Test. Method Thirty μL of the test substance was added into three single EpiDerm skin tissues, and negative control accuracy of in vitro EpiDerm Skin Irritation Test were 100%. And the results were objective, the workload was small. The issue of animal ethics was completely avoided. Conclusion In vitro EpiDerm Skin Irritation Test is the most ideal method of in vitro skin irritation.
