补肾益气平喘方对复合卵蛋白(OVA)哮喘大鼠血清IL-6及肺组织Bax、Bcl-2表达影响与地塞米松等效性随机平行对照研究 被引量:4

The Effect of Bushen Yiqi Pingchuan Prescription on IL-6 of Serum and Bax、Bcl-2 Protein of Lung Tissue in a Rat Model of Asthma

摘要 [目的]观察补肾益气平喘方对复合卵蛋白(OVA,含佐剂)致敏哮喘大鼠肺组织气道重塑及凋亡因子Bax和Bcl-2表达影响与地塞米松等效性。[方法]使用随机平行对照方法,将清洁级雄性4~5周SD大鼠32只,体质量(180±20g)按随机数字法分为4组,空白对照组、模型组、补肾益气平喘组(中药组)、地塞米松组,8只/组。复合卵蛋白(OVA,含佐剂)致敏大鼠复制哮喘模型,腹腔注射:实验第1d和第7d,各实验组腹腔注射复合卵蛋白致敏剂0.2m L,空白组等量生理盐水;喷雾激发:实验第8d各组大鼠置于超声雾化器下,1%复合卵蛋白致敏剂溶液喷雾激发,空白对照组生理盐水,30min/次·d-1,1次/d,连续14d,至实验第23d结束。灌胃干预:实验第9d,每次雾化激发前药物灌胃,中药组-补肾益气平喘方(1.35g/kg),地塞米松组-地塞米松(0.5mg/kg),连续14d,至实验第23d结束。观测大鼠行为学变化、IL-6(ELISA法),Bax、Bcl-2(Western Blot,WB法)。[结果]实验第23d(造模14d),血清IL-6空白组低于其他各组(P<0.01,P<0.05),模型组高于中药组、地塞米松组(P<0.01),中药组与地塞米松组无显著差异(P>0.05)。肺脏组织Bax模型组低于空白组(P<0.01),中药组、地塞米松组与空白组无显著差异(P>0.05),中药组、地塞米松组高于模型组(P<0.01,P<0.05),中药组与地塞米松组无显著差异(P>0.05)。Bcl2模型组高于空白组(P<0.01),地塞米松组低于空白组(P<0.01),中药组与空白组无显著差异(P>0.05);地塞米松组低于模型组(P>0.05),中药组与模型组、地塞米松组无显著差异(P>0.05)。[结论]补肾益气平喘方灌胃干预复合卵蛋白(OVA)哮喘大鼠,可抑制IL-6表达,升高Bax、Bcl-2表达,与地塞米松具有等效性。 [Objective] By observing the Bushen Yiqi Pingchuan prescription for a rat model of asthma expression of IL-6 of serum and Bax、 Bcl-2 protein of lung tissue, exploring the regulatory mechanism of Bushen Yiqi Pingchuan prescription in treating asthma preliminarily. [Methods] Using the method by intraperitoneal injection of ovalhumin(OVA) and aluminum hydroxide to make the rat mdel of asthma, the group of dexamethasone as the control, observing the effect of Bushen Yiqi Pingchuan prescription on IL-6 and Bax, Bcl-2 protein expression in a rat model of asthma. Using the ELISA method to detect the expression of IL-6, Western Blot (WB) method to detect the Bax, Bcl-2 protein expression. [Results] The model group compared with the normal group significantly enhanced IL-6 expression and IL-6 increased (P 〈0.01). After the treatment of Bushen Yiqi Pingchuan prescription, IL-6 expression decreased significantly. WB results showed that Bushen Yiqi Pingchuan Prescription can decrease the protein expression of Bcl-2 and increase the protein expression of Bax(P〈0.01). [Conclusions] Bushen Yiqi Pingchuan prescription may respectively through regulating IL-6, decreasing the protein expression of Bcl-2 and increasing the protein expression of Bax to alleviate the airway inflammation of a rat model of asthma.
