摘要
目的:建立同时测定人参各部位总皂苷水解产物中人参皂苷元原人参三醇(PPT)、人参三醇(PT)、25-羟基原人参二醇(25-OH-PPD)含量的HPLC-ELSD检测方法。方法:采用Kromasil C18柱(150 mm×4.6 mm,5μm),柱温:25℃,流速:3 m L·min^(-1);流动相为甲醇-水,梯度洗脱,ELSD检测器,气化室温度:40℃,氮气压力:2.5×10~5Pa。结果:人参皂苷PPT、PT、25-OH-PPD分别在1.6~8μg呈良好的线性关系,平均加样回收率分别为98.39%、97.97%、97.75%,RSD分别为0.94%(n=5)、1.10%(n=5)、1.27%(n=5)。结论:该方法简便、快速、重现性好,可用于人参皂苷水解产物的质量控制方法。
Objective: To establish a quantitative method for simultaneous determination of PPT, PT and 25-OH- PPD by HPLC-ELSD in different parts of Ginseng hydrolysates. Methods: A Kromasil CIs ( 150 mm × 4.6mm, 5 μm) was used to perform the determination, which was maintained at 25 ℃ throughout the analysis. Mobile phase was composed of methanol and water with flow rate at 3 mL · min^-1 under gradient elution. The evaporation light-scattering detector was used with its gasification chamber temperature at 40 ℃ and the nitrogen pressure 2. 5 ×10^5 Pa. Results: The linearity of PPT, PT, 25-OH-PPD was good in 1.6 μg - 8 μg scope, average recovery rates were 98.39%, 97.97%, 97.75%, respectively, RSD were 0. 94% (n =5) , 1.10% (n =5) , 1.27% (n =5), respectively. Conclusion: This method is simple, fast, the reproducible. The method could be used for the quality control of Ginseng hydrolysates.
