复方金银花外洗液及其单味药的体外透皮实验与抑菌活性研究 被引量:9

In vitro Antibacteriostatic Activity and Percutaneous Absorption from Flos Lonicerae Japonicae Lotion and its Water Extract

摘要 目的:观察复方金银花外洗液(外洗液)及金银花水提液(金银花液)体外透皮吸收的效果;并探讨两者对金黄色葡萄球菌、枯草芽孢杆菌、大肠杆菌、绿脓杆菌的体外抑菌活性。方法:采用改良的Franz扩散池装置,以昆明小鼠腹部皮肤为屏障,利用高效液相色谱法研究外洗液及金银花液中绿原酸的透皮吸收性能;采用管碟法测定外洗液及金银花液对4种菌的抑菌能力;用试管二倍稀释法与平板涂布法结合确定药液对4种菌的最小抑菌浓度(MIC)。结果:绿原酸在体外可以通过透皮吸收,外洗液和金银花液在8 h内绿原酸的累积透皮释药量分别为1.68、0.98μg·cm^(-2);金黄色葡萄球菌、枯草芽孢杆菌、大肠杆菌、绿脓杆菌对外洗液及金银花液均敏感;含相同绿原酸量的外洗液的抑菌圈直径都大于金银花液;含相同质量浓度的绿原酸和生药的外洗液对4种菌的MIC值均比金银花液对4种菌的MIC值小。结论:外洗液及金银花液均有体外透皮吸收作用和抑菌活性,且复方的最高安全剂量和功效要强于单味药。 Objective:To detect the bacteriostatic activity of Flos Lonicerae Japonicae lotion and its water extract on Staphylococcus aureus,Bacillus stubtilis,Escherichia coli and Pseudomonas aeruginosa in vitro.Methods:The minimum inhibitory concentration (MIC) was scaled by tube dilution and plate smearing methods.The percutaneous absorption potential of their chlorogenic acid were detected by HPLC,in which via mouse’s abdominal skin cell viability was determined using Flos Lonicerae Japonicae lotion and its water extract to the four bacteria.Results: Chlorogenic acid could be absorbed via skin in vitro,so after 8 hours treatments of the Flos Lonicerae Japonicae lotion and its water extract,the chlorogenic acids’ amount were 1.68 and 0.98 μg·cm^-2,respectively.Staphylococcus aureus,Bacillus stubtilis,Escherichia coli and Pseudomonas aeruginosa were sensitive to Flos Lonicerae Japonicae lotion and its water extract.Meanwhile,the inhibition zone diameter of Flos Lonicerae Japonicae lotion was bigger than that of its water extract,but the lotion’s MIC was less than water extract’s.Conclusion: Both Flos Lonicerae Japonicae lotion and its water extract can be used as antibiotic treatment and absorbed via skin in vitro.Moreover,the effect of compound of Flos Lonicerae Japonicae was better than individual drug.
