慈姑多糖和慈姑水提部位对异烟肼和利福平合用致肝细胞损伤保护作用的比较研究 被引量:7

摘要 目的比较慈姑多糖和慈姑水提部位在体外对异烟肼(INH)和利福平(RFP)合用致肝细胞损伤的保护作用。方法采用系统化学溶剂萃取法制备慈姑水提部位,采用水提醇沉法制备慈姑多糖。培养Hep G2肝细胞,建立异烟肼和利福平合用(INH/RFP)致体外肝细胞损伤模型,给予慈姑水提部位或慈姑多糖处理后,利用MTT法检测肝细胞的存活率,并检测细胞培养上清液中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、乳酸脱氢酶(LDH)、丙二醛(MDA)的浓度。结果与模型组比较,慈姑水提部位组和慈姑多糖组均可增加细胞存活率(P<0.05),降低INH/RFP损伤肝细胞培养上清液中ALT、AST、LDH、MDA水平(P<0.05),慈姑多糖组较慈姑水提物部位组细胞存活率更高,细胞培养上清液中ALT、AST、LDH、MDA水平更低。结论慈姑多糖和慈姑水提部位对体外INH/RFP致肝细胞损伤均有保护作用,慈姑多糖作用更强,其作用可能与抗脂质过氧化有关。 Objective To compare the protective effect of polysaccharides and aqueous extracts of Sagittaria Sagittifolia against hepatocellular injury caused by isoniazid and rifampin co-administration. Methods Aqueous extracts of Sagittaria Sagittifolia were prepared by using chemical solvent extraction system. Polysaccharides of Sagittaria Sagittifolia were prepared by the method of water extraction and alcohol precipitation. The model of hepatocellular injury in vitro caused by isoniazid and rifampin co-administration was achieved by cultivating HepG2 type of hepatocyte. After treated with aqueous extracts or polysaccharides of Sagittaria Sagittifolia, hepatocyte viability was detected by MTF-assay. Meanwhile, the concentration of alanine aminotransferase (ALT), Aspartate aminotransferase ( AST), lactic dehydrogenase (LDH) and malondialdehyde (MDA) in cell culture supernatants was detected. Results Compared with the model group, the hepatocyte viability became higher( P 〈 0.05 ) , while the concentration of ALT, AST, LDH and MDA in cell culture supernatants was lowered ( P 〈 0. 05 ) in both groups of aqueous extracts and polysaccharides. Compared with the aqueous extracts group, the hepatocyte viability was much higher, while the concentration of ALT, AST, LDH and MDA in cell culture supernatants was much lower in the group of polysaccharides. Conclusion Both aqueous extracts and polysaccharides of Sagittaria Sagittifolia have the protective effect against hepatocellular injury caused by isoniazid and rifampin co-administration in vitro, while the effect of the latter may be stronger. Such protective effect may be related to the mechanism of anti-lipid-peroxidation.
