槲皮黄酮对急性缺血性卒中大鼠的保护作用及机制 被引量:3

摘要 目的探讨槲皮黄酮对急性缺血性卒中大鼠的保护作用及机制。方法健康成年雌性SD大鼠35只,随机分为假手术组、缺血再灌注模型组、低、中、高剂量榭皮黄酮治疗组(2、4、8 mg/kg)。采用TTC染色法检测大鼠脑梗死面积,ELISA试剂盒检测脑脊液中炎症因子IL-6、TNF-α表达水平,Western blot技术检测TRAF6/IKKβ/NF-κB通路蛋白表达水平。结果与缺血再灌注组比较,中、高剂量槲皮黄酮处理可明显减轻缺血再灌注所致的大脑梗死面积,降低脑脊液中炎症因子IL-6、TNF-α的表达,抑制TRAF6、p-IKKβ的表达及NF-κB的激活转位,差异均有统计学意义(P<0.05)。结论槲皮黄酮对急性缺血性卒中大鼠具有一定的保护作用,其机制可能与抑制TRAF6/IKKβ/NF-κB信号通路有关,进一步减轻炎症损伤。 Objective To investigate the protective effect and mechanism of quercetin on acute ischemic stroke in rats. Methods Thirty-five female SD rats were randomly divided into five groups, including the sham group,the ischemic/reperfusion group, three different doses of quercetin groups. Infarct area in rat brain was determined via TTC examination. Inflammatory factors IL-6 and TNF-α in cerebrospinal fluid were measured using ELISA Kit. The proteins involved in the TRAF6/IKKβ/NF-κB signaling were determined using western blot. Results Compared with the ischemic/reperfusion group,quercetin attenuated the ischemic/reperfusion-induced cerebral infarct area. Inflammatory cytokines IL-6 and TNF-α in the cerebrospinal fluid were decreased in mild and high doses of quercetin groups. Quercetin significantly inhibited the expression of TRAF6 and p-IKKβ, and redistribution of NF-κB( P〈0. 05). Conclusion Quercetin may have a protective effect in acute ischemic stroke rats through inhibiting the TRAF6/IKKβ/NF-κB signaling.
