摘要
建立了生物酶转化液中β-丙氨酸的高效液相色谱检测方法。优选氨基键合硅胶柱;流动相采用0.05 mol/L磷酸二氢钾∶乙腈(35:65),流速1.0 ml/min,波长205 nm,柱温30℃,配合对照溶液,实现转化液中β-丙氨酸含量的标定。检测表明:线性回归相关度0.999 6,回收率大于99.49%,实验重复性RSD为0.15%(n=6),实验稳定性RSD为1.55%(n=4)。方法简单易行,分离效果好,解决了传统方法在转化液成分复杂时测量结果偏差较大的瓶颈。
A methodology of high performance liquid chromatography (HPLC) is developed for the determination of β-alanine in bio-enzyme transformation solution. Amino bonded silica gel column is optimized selected; flow phase of 0.05 mol/L potassium dihydrogen phosphate : acetonitrile (35:65) is adopted with flow rate of 1 ml/min, wavelength 205 nm, column temperature 30 Celsuis, to realize the content calibration of β-alanine in transformation solution along with the contrast solution. The results show that, the linear regression correlation is 0.999 6, the recovery rate is more than 99.49%,the repeatability of RSD is 0.15% (n=6), and the stability of RSD is 1.55% (n=4). The methodology has advantages of simple operation and good separation effect, and solves the bottleneck of large deviation of measurement result in traditional methods when the composition of transformation solution is complex.
