川芎挥发油鼻腔喷雾剂对脑缺血大鼠急性期的治疗作用

Therapeutic Effects of Chuanxiong Rhizome Volatile Oil Nasal Spray on Acute Cerebral Ischemia in Rats

目的:探讨川芎挥发油鼻腔喷雾剂对脑缺血/再灌注脑损伤大鼠的保护作用及其可能的作用机制。方法:采用Longa线栓法制备大脑中动脉阻塞脑缺血大鼠模型,设立模型对照组、川芎挥发油生理盐水口服给药组、川芎挥发油喷雾剂组(1:1)、川芎挥发油喷雾剂剂量递增组(剂量从1:20至1:1)。用药24h后观察大鼠一般状况和神经行为学、脑缺血梗死体积及局部脑血流量的变化,测定脑组织中SOD、MDA、LDH的含量,并进行病理学组织检查。结果:川芎挥发油喷雾剂能显著地延长大鼠缺血的存活时间;改善大鼠脑缺血造成的行为缺陷;缩小脑梗死的体积;川芎挥发油喷雾剂能剂量依赖性地提高局部脑血流量并增加局部运动血细胞数量;川芎挥发油喷雾剂还能提高脑组织中SOD、LDH活性,减少MDA含量;病理组织检查显示,川芎挥发油喷雾剂能改善脑缺血大鼠神经细胞损伤,减少组织坏死。结论:川芎挥发油鼻腔喷雾剂对脑缺血具有明显的保护作用,是一个值得推荐及研究的给药途径。

Objective ： To observe the protective effects of Chuanxiong rhizome volatile oil nasal spray on cere- bral isehemia- repeffusion injury in rats and explore its mechanism. Methods： The cerebral ischemia model of middle cerebral artery occlusion was established with Longa suture method in rats. Four groups were set up in the study, they were control group, Chuanxiong rhizome volatile oil administered by the oral route with saline group, Chuanxiong rhizome volatile oil spray group （ doses of 1 ： 1 ） , Chuanxiong rhizome volatile oil spray dose - increase group （ doses from 1：20 - 1 ： 1 ）. After 24 hours of administration, the general condition, neural behavior, ischemic cerebral infarction volume and the regional cerebral blood flow were detected respectively. The content of SOD, MDA,LDH were determined and the pathological change of cerebral tissues were observed. Results： Chuanxiong rhizome volatile oil spray can evidently prolong survival time of rats in the states of anoxia and ameliorated rats＇ neurologic deficit. The volume of cerebral infarction in Chuanxiong rhizome volatile oil spray group was smaller than that of control group. Chuanxiong rhizome volatile oil spray, in a dose - dependent manner, improve the regional cerebral blood flow and increase the number of moving blood cells. Chuanxiong rhizome volatile oil spray could also increase the activities of SOD and LDH, reduce the content of MDA. The pathological examination showed that Chuanxiong rhizome volatile oil spray could significantly protect brain from ischemia, reperfusion injury, reduce the necrosis of neuron and the cerebral infarction. Conclusion： Chuanxiong rhizome volatile oil nasal spray has a obvi- ously protective effect against ischemia brain damage and is valuable to be further studied as a new preparation .
