复方黄精茶对糖尿病大鼠糖脂代谢的影响及血管保护作用

Protective effect on glycolipid metabolism and blood vessels of diabetic rats from compound polygonatum sibiricum tea

目的研究复方黄精茶对2型糖尿病模型大鼠的治疗效果,了解复方黄精茶对糖脂代谢的影响及血管的保护作用。方法 40只实验大鼠,随机取10只大鼠作为正常对照组,正常饮食;其余大鼠高脂高糖饮食30 d后,用链脲佐菌素(STZ)加高脂高糖饲料诱导2型糖尿病大鼠模型,造模成功后随机分为糖尿病模型组、二甲双胍组[300 mg/(kg·d)]及复方黄精茶组[生药量5 g/(kg·d)],每组10只,灌胃给药8周后,测定各组糖脂代谢指标、HE染色法检测大鼠胸主动脉病理学形态改变,取各组大鼠肾动脉制成肾动脉环,比较各组乙酰胆碱(ACh)诱导的舒张作用。结果与糖尿病模型组比较,复方黄精茶显著降低糖尿病大鼠空腹血糖、糖化血红蛋白、血糖曲线下面积水平,改善总胆固醇、甘油三酯、高密度脂蛋白、低密度脂蛋白等脂代谢指标,及减轻血管内皮增厚,减少弹力纤维蛋白断裂现象,增强ACh诱导的肾动脉舒张效应。结论复方黄精茶能改善糖尿病大鼠糖脂代谢及保护血管。

Objective To study the treatment effect type 2 diabetes,and understand its protective effect on of compound polygonatum sibiricum tea on rats of glycolipid metabolism and blood vessels of diabetic rats. Methods Among totally 40 rats, 10 were randomly selected as the control group which was given the normal diets ,while the other rats were given high fat and high sugar diets for 30 days then were set up type 2 diabetic rats model induced by streptozotocin（STZ） and feedstuff with high fat and high sugar. After that, they were divided into model group, metformin group [ 300 mg/（kg .d） ] and compound polygonatum sibiri- cure tea group[with 5 g/{kg .d） crude drug], 10 in each group. After they were given by garage for 32 days, the quotas of glycolipid metabolism of each group were measured, pathological changes of thoracic aor- ta in rats were measured by HE staining method and renal arteries from each rat group were taken to made renal artery rings for studying their vasodilator effects induced by acetylcholine （ACh）. Results Compared with model group,compound polygonatum sibiricum tea significantly reduced the fasting blood-glucose, gly- cosylated hemoglobin and the area under the curve of blood glucose level, improved the quotas of glycolipid metabolism such as total cholesterol, triglyceride{ TG）, high density lipoprotein, low density lipoprotein and lightened the thickening of vascular endothelial structure and the rupture of elastic fiber protein. What＇ s more,it enhanced the vasodilator effects of renal artery induced by Ach in diabetic rats. Conclusion Com- pound polygonatum sibiricum tea can improve glycolipid metabolism and protect blood vessel in diabetic rats.
