摘要
目的:观察缺氧缺血对新生大鼠脑组织MMP-9、TIMP-1含量的影响及红景天苷的干预作用。方法:选择健康7日龄清洁级Wistar大鼠120只,建立缺血缺氧模型,分为假手术组、HIBD组、HIBD+红景天苷组,分别给予相应药物,于6h、1d、2d、5d、14d时间点取材,运用免疫组织化学法检测MMP-9、TIMP-1含量。结果:HIBD组大鼠MMP-9、TIMP-1水平显著高于假手术组(P<0.05)。经红景天苷干预后,HIBD+红景天苷组大鼠MMP-9水平显著降低,6h、1d、2d时含量与HIBD组大鼠比较差异均具有统计学意义(P<0.05);TIMP-1水平显著降低,1d、2d、5d时含量与HIBD组大鼠比较差异均具有统计学意义(P<0.05)。结论:降低脑组织MMP-9、TIMP-1含量可能是红景天苷发挥神经保护作用的机制之一。
Objective :To observe intervention of Rhodiola on MMP-9/TIMP-1 system of neonatal rat brain tissue with hypoxic ischemic. Methods:selected healthy 7 clay old newborn Wistar rats, 120 rats were divided into three groups: control group, the HIBD group, HIBD + drug groups, each group was tested at 6h, 1d and 2d, 5d, 14 d with immunohistochemical method on the MMP-9 and TIMP-1. Results:the level of MMP-9 HIBD group in 6h,1d, 5d were significantly higher than control group (P〈 0.05), HIBD+ Rhodiola group reduced expression of MMP-9 of 6h, ld and 2d, and there were significant differences (P〈0.05) ; Expression of TIMP-1 on 1d,2d,5d corresponding decrease, and there were significant differences (P〈0.05). Conclusion: The steady state reconstruction of brain tissue on MMP-9/TIMP 1 system may be one of the mechanisms about protective effection of rhodiola glucoside in HIBD.
