全血细胞嗜乳脂蛋白样3基因表达在支气管哮喘发病中作用的研究 被引量:1

Role of expression levels of BTNL3 mRNA in global blood cells in pathogenesis of bronchial asthma

摘要 目的 探讨全血细胞嗜乳脂蛋白样3 (BTNL3)基因表达在支气管哮喘 (简称哮喘) 发病中的作用机制。方法 采用实时荧光定量 PCR技术检测65例哮喘患者和26例健康志愿者全血细胞 BTNL3mRNA 表达水平,采用酶联免疫吸附法检测其血浆IL-4浓度,并分析两者之间的相关性。结果 对照组、轻度持续组、中度持续组、重度持续组 BTNL3 mRNA 相对表达量为:0.63±0.2、 0.47±0.15、0.44±0.19、0.34±0.19;方差分析和t检验显示,对照组 BTNL3mRNA 表达高于哮喘各组,并且轻度持续组表达高于中度持续组,中度持续组表达高于重度持续组 (P 值均<0.05)。相关分析显示,全血细胞 BTNL3mRNA 相对表达量和血浆IL-4浓度具有负线性相关性 (Pearson相关系数=-0.857,P <0.01)。结论 全血细胞 BTNL3mRNA 的表达水平和哮喘的严重程度呈负相关,并与血浆IL-4浓度呈负相关,提示BTNL3可能通过抑制Th2免疫反应降低哮喘的发病和严重程度。 Objective To investigate the role of expression levels of butyrophilin like protein 3 (BTNAL3) mRNA in global blood cells in pathogenesis of bronchial asthma(asthma). Methods Sixty- five asthma patients with diverse severities were recruited in the study. Twenty-six healthy volunteers were recruited as control. The expression levels of BTNL3 mRNA were detected by real time quantitative polymerase chain reaction (PCR), the serum interleukin-4 (IL-4) levels were detected by enzyme-linked immunosorbent assay (ELISA). The results were compared among groups and the correlation analysis were performed between the expression levels of BTNL3 mRNA and serum IL-4 level. Results The relative expression levels of BTNL3 mRNA in control group, mild asthma group, medium asthma group and severe asthma group were 0.63±0.2,0.47±0.15,0.44±0.19,0.34±0.19. The relative expression levels of BTNL3 mRNA in control group was higher than asthma groups, and mild asthma group was higher than medium asthma group, medium asthma group was higher than severe asthma group ( P 〈 0.05). The correlation analysis showed that the BTNL3 mRNA expression were negative correlated with serum levels of IL-4 ( r = -0.857, P 〈0.01). Conclusions The BTNL3 mRNA expression in global blood cells were negative correlated with the asthma severitiy, and were negative correlated with the serum levels of IL-4. This results recommended that the BTNL3 gene may have an inhibitional effect on the Th2 immunoreaction,and then reduce the morbidity and the severitiy of asthma.
