牡丹‘璎珞宝珠’不定芽增殖及生根诱导研究 被引量:1

摘要 以牡丹‘璎珞宝珠’为材料,对影响其不定芽分化、生根的因素进行了研究,并对根源基的解剖学结构进行了观察.结果表明:在不定芽的增殖培养中,分化较好的培养基为MSB+1.5mg·L^(-1)6-BA+0.05mg·L^(-1)NAA,不定芽再生率最高为(90.30±1.56)%,增殖系数最高为5.91±0.20;诱导生根最佳培养基为1/2MSB+3.0mg·L^(-1)IBA+0.5mg·L^(-1) NAA+1g·L^(-1)活性炭(AC),生根率最高达到(75.25±2.43)%,生根量为3.30±0.09,根长度最长达到(1.40±0.11)cm,与其他处理差异显著.牡丹组培苗的不定根属于诱生根原始体型,不定根原始体起源于维管束的形成层细胞,继代次数与生根有相关性.前期4℃低温处理7d培养有利于生根,自然散射光照射下,生根状况最好,根的平均长度为(2.62±0.28)cm,移栽成活率达(85.4±1.13)%. 'yingluobaozhu' peony(Paeonia suffruticosa)was used as material in this study.The factors of affecting the adventitious bud differentiation and rooting were studied,and the anatomical structure of root primordium was observed.The results showed that the better medium for adventitious buds differentiation was MSB+6-BA 1.5mg·L^(-1)+NAA 0.05mg·L^(-1).The highest adventitious buds regeneration rate was(90.30±1.56)%,and the highest proliferation coefficient was 5.91±0.20.The best rooting medium was 1/2MSB+IBA 3.0mg·L^(-1)+ NAA 0.5mg·L^(-1)+1g·L^(-1) activated carbon(AC),in which penoy's rooting rate was(75.25±2.43)%,its rooting amount is 3.30±0.09 and its longest root length reached(1.40±0.11)cm.This was significantly different from other treatments(P<0.05).The adventitious roots of penoy suffruticosa tissue culture seedlings belonged to the induced root of original shape.Adventitious roots primitive body originated in the vascular cambium cells,the number of secondary culture was correlated with rooting.The 4℃ treatment for 7dwas beneficial to rooting early,and the root grew best in the natural light scattering.Its average length of root was(2.62±0.28)cm,and the survival rate of transplanting was(85.4±1.13)%.
