摘要
目的:探讨双藤痹痛凝胶膏剂对胶原诱导性关节炎模型(collagen-induced arthritis,CIA)大鼠滑膜组织中炎性细胞因子表达的影响,探讨其治疗作用机理。方法:Wistar大鼠40只,随机分成4组,分别为正常组、模型对照组、双藤痹痛凝胶高、低剂量组,通过建立CIA大鼠模型,采用酶联免疫吸附测定法(enzyme-linked immunosorbent assay,ELISA)检测血清中肿瘤坏死因子-α(TNF-α)、白细胞介素(IL)-1β及IL-6的含量,逆转录-聚合酶链式扩增反应(Reverse Transcription-Polymerase Chain Reaction,RT–PCR)法测定滑膜组织中TNF-α、IL-1β及IL-6 mRNA的表达水平。结果:ELISA法检测结果显示,与模型组比较,双藤痹痛凝胶高、低剂量组大鼠血清中TNF-α、IL-1β及IL-6?的含量显著下降(P<0.05或P<0.01);RT–PCR检测显示,与模型组比较,双藤痹痛凝胶膏剂高、低剂量组大鼠滑膜组织中TNF-α、IL-1β及IL-6 mRNA的表达明显降低。结论:双藤痹痛凝胶膏剂对胶原诱导性关节炎大鼠有一定的治疗作用,其机制可能与降低血清及滑膜组织中TNF-α、IL-1β及IL-6水平有关。
Objective: To study the effect of Shuangteng Bitong Cataplasm on inflammatory cytokines in rats with collagen-induced arthritis. Methods: Forty Wistar rats were randomly divided into 4 groups,normal control group,model control group,high-dose and low-dose Shuangteng Bitong Cataplasm group. Rats' arthritis was induced by collagen. The content of TNF-α,IL-1β and IL-6 in serum were evaluated by ELISA,and the expression levels of TNF-α,IL-1β and IL-6 in synovial tissues were evaluated by RT –PCR. Results: ELISA assay showed: compared with the model groups,the content of TNF-α,IL-1β and IL-6 in serum was decreased remarkably in the low and high-dose of Shuangteng Bitong Cataplasm groups.( P < 0. 05 or P < 0. 01),and RT – PCR indicated: the expression levels of TNF-α,IL-1β and IL-6 in synovial tissues was decreased remarkably in the low and high-dose of Shuangteng Bitong Cataplasm groups. Conclusion: Shuangteng Bitong Cataplasm has a certain protective effect on CIA rats and its mechanism is possibly relevant with the down-regulation of the expressions of TNF-α,IL-1β and IL-6 mRNA.
