摘要
探讨适合蛆症异蚤蝇幼期的石蜡切片制作方法,并对其幼期内部主要组织结构进行了观察.结果显示,Carnoy固定液(V(无水乙醇)∶V(氯仿)∶V(冰醋酸)=6∶3∶1)适于蛆症异蚤蝇幼期的固定.将蛹头部或整体蛹壳剥离,有利于固定、脱水和包埋,蛹需过夜浸蜡.在切片上可清楚的显示3龄幼虫的口器、唾液腺、脂肪体、中肠以及36h蛹的脑组织、复眼、卵巢和腹部肌肉等主要内部结构.
The proper method of preparing the paraffin section of immatures of Megaseliascalaris（Loew）is discussed,and their internal tissue structures are observed.The results show that Carnoy fixative（V（alcohol）∶V（chloroform）∶V（acetic acid）=6∶3∶1）is suitable to fix the immatures.The head or the shell of the whole body of pupa should be peeled off,which is helpful for fixation,dehydration and embedding of pupa.Pupa needs overnight paraffin.The main internal structures,such as the cephalo skeleton,salivary glands and midgut of 3instar larva,and brain tissue,compound eye,ovary and abdomen muscles of 36 hpupa could be clearly observed on the slices.
