摘要
目的观察叶下珠提取物对人肾癌786-0细胞增殖抑制作用并探讨叶下珠提取物发挥该作用的机制,为叶下珠的临床应用更进一步的提供理论依据。方法用不同浓度的叶下珠(浓度分别为1、10、100、1000μg/m L)作用于人肾癌786-0细胞株,采用CCK-8法检测叶下珠对肾癌786-0细胞增殖效应的影响,采用流式细胞术(FCM)检测叶下珠对肾癌786-0细胞周期的影响,采用蛋白印迹法检测叶下珠对Survivin蛋白表达水平的影响。结果 CCK-8法结果提示,不同浓度的叶下珠均可显著抑制肾癌786-0细胞的增殖,且抑制率呈时间、剂量依赖关系。FCM检测发现叶下珠可使肾癌786-0细胞周期中的G0/G1期比例增加,S期缩短。蛋白印迹法显示,叶下珠可使肾癌786-0细胞中Survivin蛋白表达水平降低。结论通过建立人肾癌786-0细胞的体外抗肿瘤实验模型,预试验结果表明叶下珠乙醇提取物对人肾癌786-0细胞增殖具有抑制作用。
Objective To observe the effect of Phyllanthus extract on proliferation of human renal cell carcinoma 786-0 cells, and to investigate the mechanism of the inhibitory effect of Phyllanthus extract, and to provide theoretical basis for further clinical application. Methods Different concentrations of Phyllanthus urinaria(concentrations of 1, 10, 100, 1000 g/m L) in human renal cell carcinoma 786-0 cell line, CCK-8 method was used to detect the effect of Phyllanthus effect on proliferation of renal cell carcinoma 786-0 cells, using flow cytometry(FCM) detection effect of Phyllanthus on renal cell carcinoma 786-0 cell cycle effect of Phyllanthus,detection on the expression of Survivin protein by Western blot. Results The results of CCK-8 showed that different concentrations of Phyllanthus significantly inhibited the proliferation of 786-0 cells, and the inhibition rate was time and dose dependent. FCM detected that Phyllanthus can make renal carcinoma 786-0 cell cycle in G0/phase G1 was increased, and S shortened. Western blot showed the level of p.urinaria, reduces the expression of Survivin protein in renal cell carcinoma 786-0 cells. Conclusion The establishment of experimental model of human renal cell carcinoma 786-0 cells in vitro anti pre test results showed that the ethanol extract of Phyllanthus has inhibitory effect on the proliferation of human renal cell carcinoma 786-0 cells.
