地高辛对非霍奇金淋巴瘤细胞体外药物敏感性的研究 被引量:1

摘要 目的了解强心苷类药物地高辛对非霍奇金淋巴瘤组织细胞的体外药物敏感度及与阿糖胞苷联合作用效果。方法体外培养非霍奇金淋巴瘤细胞,予不同浓度地高辛、阿糖胞苷及两药联合进行干预,应用MTS比色法检测细胞的生长及凋亡状况。结果(1)地高辛对非霍奇金淋巴瘤细胞的体外药物敏感度:①各浓度组地高辛分别作用于12组非霍奇金淋巴瘤细胞24 h、48 h和72 h后,各组抑制率均不同程度增高;②不同浓度地高辛对非霍奇金淋巴瘤组织细胞的作用敏感率随时间变化差异有显著性。(2)地高辛对阿糖胞苷化疗效果的协同作用:①析因设计方差分析显示:不同浓度的地高辛对非霍奇金淋巴瘤细胞的作用有显著性差异(F_(地高辛)=20.567,P<0.01),不同浓度的阿糖胞苷作用组亦有显著性差异(F_(阿糖胞苷)=57.182,P<0.01)。且两者具有一定协同作用。②单独效应方差结果分析显示:各浓度组地高辛对非霍奇金淋巴瘤细胞的作用仅在阿糖胞苷浓度为100μmol/L时,差异具有显著性(F=24.390,P<0.01)。结论(1)地高辛在体外可对非霍奇金淋巴瘤细胞有一定抑制作用。(2)地高辛对阿糖胞苷的化疗效果有一定增强,在阿糖胞苷高浓度时协同作用效果更为明显。 Objective In this study,non-Hodgkin's lymphoma cells were cultured in vitro,then treated with digoxin or/and cytosine arabinoside,observe the digoxin sensitivity and explore the synergy effect combined with cytosine arabinoside.Methods Non-Hodgkin' s lymphoma cells were cultured in vitro and treated with different concentrations of digoxin,or digoxin and cytosine arabinosidecombination.Cell inhibition rate was investigated by MTS assay.Results 1.Digoxin sensitivity on non-Hodgkin's lymphoma in vitro;①After non-Hodgkin's lymphoma cells treated with different concentrations of digoxin for 24 h,48h and 72 h,the inhibition rates were increased at different degree.The sensitivity of the NHL cells treated with 25nmol/L Digoxin for 24 h was the worst,treated with 200nmol/L Digoxin for72 h was the best.②After NHL cells treated with different concentration of digoxin for 24h、48h and 72 h,the average inhibition rate increased at different degree,with statistical significance(P〈0.05) when treated with different concentration of digoxin for 72 h,but not for treated for 24 h and 48h(P〉0.05).③Through Nemenyi test for 72 h,we found only difference between 25nmol/L group and 200nmol/L had statistical significance(P〈0.05).Therefore we suspect the inhibition of non-Hodgkin' s lymphoma cells treated by higher concentration of digoxin was more apparent.2.Sensitizing effect of digoxin combined with cytosine arabinoside in non-Hodgkin lymphoma cells:①After NHL cells treated with digoxin combined with cytosine arabinoside for 48 h,the average inhibition rate increased with the concentration increased.②The result showed that;the effect of cytosine arabinoside had statistical significance when the concentration of cytosine arabinoside was 100 μmol/L.Conclusions 1.The survival of NHL cells was inhibited by digoxin in vitro with a concentration dependent manners.2.Digoxin has the effect of chemosensitization to cytosine arabinoside on NHL cells in vitro.Application of small dose cytosine arabinoside with digoxin can achieve the same effect of conventional dose cytosine arabinoside chemotherapy.
