The Effect of Matrix Metalloproteinase-2 Gene Silence in the Inhibition of Invasion and Growth of Laryngeal Cancer Cells

摘要 目的探讨重组慢病毒介导的基质金属蛋白酶-2(MMP-2)沉默对喉鳞癌侵袭生长的抑制作用。方法该研究于2016年1—3月,采用RNA干扰技术,运用MMP-2基因si RNA的重组慢病毒(MMP-2-RNAi-Lentivirus)转导喉鳞癌Hep-2细胞,通过侵袭小室实验观察MMP-2-RNAi-Lentivirus转染后喉癌细胞侵袭能力的变化;构建喉鳞癌动物模型时,18只BALB/c Nude裸鼠肿瘤生长直径达到1.0 cm时随机分组,治疗组9只,空载体对照组9只,给予瘤内注射等量的GFP-Lentivirus,进一步观察其对喉癌侵袭转移的影响。结果侵袭实验显示,实验组Hep-2细胞穿过人工基底膜的细胞数为(12±4)个,明显少于空载体对照组的(35±6)个,差异具有统计学意义(P<0.05);体内实验显示,MMP-2-RNAi-Lentivirus瘤内注射后,裸鼠移植瘤平均重量为(1.186±0.225)g,平均体积为(0.974±0.216)cm^3,对照组分别为(2.127±0.344)g、(1.618±0.272)cm^3,实验组低于对照组,差异具有统计学意义(P<0.05)。结论重组慢病毒介导的MMP-2基因沉默能有效的抑制喉癌细胞的增殖过程,可为喉癌基因治疗提供实验参考。 Objective To investigate the inhibitory effect of recombinant slow virus mediated MMP-2 silencing on laryngeal squamous cell carcinoma. Methods From January 2016 to March 2016,by RNA interference technology,with MMP-2 si RNA MMP-2-RNAi-Lentivirus gene transduction of laryngeal squamous cell carcinoma Hep-2 cells,laryngeal cancer cell invasion ability was observed after transfection,by constructing the laryngeal carcinoma xenograft model,18 Nude BALB/c nude mice were randomly divided into 1.0 cm,the treatment group was 9,and the control group was 9,and the amount of GFP-Lentivirus was injected into the tumor. The effect of the treatment on the invasion and metastasis of laryngeal carcinoma was further observed. Results The invasion assay showed that the number of cells in the experimental group Hep-2 cells through the artificial basement membrane were(12±4),significantly less than the empty vector control group(35 ±6),the difference was statistically significant(P 〈 0.05); in vivo,intratumoral injection of MMP-2-RNAi-Lentivirus,the average weight of nude mice for(1.186±0.225)g and the average volume(0.974±0.216)cm3,the control group were(2.127±0.344)g,(1.618±0.272)cm3,the experimental group was lower than the control group,the difference was statistically significant(P 〈 0.05). Conclusion The recombinant virus mediated MMP-2 gene silencing can effectively inhibit the proliferation of laryngeal cancer cells,which can provide experimental reference for gene therapy of laryngeal cancer.
