短葶山麦冬多糖质量控制方法研究 被引量:4

摘要 目的建立短葶山麦冬多糖质量控制方法。方法采用高效凝胶渗透色谱(HP-GPC)法测定短葶山麦冬多糖相对分子质量及其分布和样品中多糖含量。色谱条件:Ultrahydrogel 250(300mm×7.8mm,5μm)凝胶色谱柱,流动相为水,流速0.5m L·min^(-1),柱温35℃,进样量20μL,检测器为示差检测器。结果短葶山麦冬多糖的主峰出现在16.23~18.75min,重均分子量约为1831~1910Da,分子量分布系数1.31~1.35;在0.01~2.0mg·m L^(-1)呈良好的线性关系(r=0.9991);短葶山麦冬多糖的含量约为80%(以葡聚糖标准品(Fluka)为对照)。多糖检测限和定量限分别为0.1μg和0.2μg。结论所建立的方法简单、快速、准确、重现性好、灵敏度较高,可作为短葶山麦冬多糖质量控制的方法。 ABSTRACT: OBJECTIVE To develop a method of quality control of Liriope muscari (Decne) Baily polysaccha- rides. METHODS The method was that using HP-GPC detected the relativemolecular mass, molecular mass distri- bution of Liriope muscari (Decne) Baily polysaccharides and contents of polysaccharides in samples. The separations was carried out 0. 5mL · min - 1 tor. RESULTS with Ultrahydrogel 250(300mm×7.8mm,5μm)column,water as the mobile phase at flow rate , the column temperature at 35℃, the injection volume 20μL, and differential index detec- muscari(Decne) Baily polysaccharides peaks were located within 16.23 - 18.75min, the weight averagemolecular mass were 1831 - 1910 Da, molecular mass distribution coefficient were 1.31 - 1.35, the linearity was good in the range of 0. 01 - 2. 0mg · mL-1 (r = 0. 9991 ) , and the content of Liriope muscari (Decne) Baily polysaccharides were about 80% (contrast with dextran standards Fluka), the detection and quantitation limit were 0. 1 μg and 0. 2μg. CONCLUSION This method is simple, quick, reproducible and accurate, which could be uesd for quality control of Liriope muscari (Decne) Baily polysaccharides.
