摘要
目的 对射干 Belamcanda chinensis(L.) DC.和鸢尾 Iristectorum Maxim.的染色体数目和核型进行了研究。方法 用饱和对二氯苯溶液 ,卡诺氏固定液及盐酸处理 ,水解 ,用改良苯酚品红染色液染色、压片镜检。结果 射干的染色体数目为 2 n=32 ,核型公式为 2 n=32 =16 m+12 sm+4 st,核型类型为 2 B;鸢尾的染色体数目为 2 n=2 8,核型公式为 2 n=2 8=10 sm+18sm,核型类型为 2 B。
Objective The chromosome karyotypes of Belamcanda chinensis(L.)DC and Iris tectorum Maxim were studied.Method Root-tips of two kinds of samples were treated in saturated para-dichlorobenzene solution for 3~5 h,fixed for 24 h with Carnoy's fixative,differentiating for 15 h with 1 mol/L hydrochloric acid at 60 ℃,staining ,pressing to section,observing them under microscope.Results Chromosome number of Belamcanda chinensis(L.)DC is 2 n=32 , with 2B karyotype, the karyotype fomula is 2 n=32=l6 m+12 sm+4 st; The chromosome number of Iris tectorum Maxim is 2 n=28,with 2 B karyotype ,and the karyotype formula is 2 n=28=10 sm+18 sm.Conclusion There were significant differences in the numbers and karyotypes of somatic cell chromosome between Belamcanda chinensis(L.)DC and Iris tectorum Maxim.
