子痫前期胎盘组织中LAMA4蛋白表达量及其与滋养细胞侵袭、胎盘缺氧的相关性 被引量:6

Expression level of LAMA4 protein in placental tissue of preeclampsia patients and its correlation with invasion of trophoblast cells and placental hypoxia

摘要 目的探讨子痫前期胎盘组织中层粘连蛋白α4链(LAMA4)蛋白表达量及其与滋养细胞侵袭、胎盘缺氧的相关性。方法收集2012年5月-2016年5月在该院接受治疗及分娩的子痫前期患者56例作为观察组;另选取同期进行产检及分娩的正常产妇50例作为正常对照组。根据胎盘组织中LAMA4蛋白表达中位数,观察组患者被分为高LAMA4蛋白组和低LAMA4蛋白组,每组各28例。采用Western-Blot法检测胎盘组织中LAMA4蛋白表达量;采用荧光定量PCR法检测胎盘组织中滋养细胞侵袭相关基因mRNA表达量;采用化学发光法检测脐血中胎盘缺氧相关指标含量。结果观察组胎盘组织LAMA4蛋白表达量低于正常对照组(P<0.05);高LAMA4蛋白组、低LAMA4蛋白组胎盘组织中基质金属蛋白酶-9(MMP-9)、血管内皮生长因子(VEGF)、胎盘生长因子(PLGF)mRNA表达量低于正常对照组,可溶性fms-样酪氨酸激酶受体-1(sFlt1)mRNA表达量高于正常对照组,脐血胎盘缺氧指标,缺氧诱导因子-1α(HIF-1α)、缺氧诱导因子脯氨酸羟化酶-1(HPH-1)、瘦素含量均高于正常对照组(P<0.05);高LAMA4蛋白组胎盘组织中MMP-9、VEGF、PLGFmRNA表达量高于低LAMA4蛋白组,sFlt1mRNA表达量低于低LAMA4蛋白组,脐血胎盘缺氧指标HIF-1α、HPH-1、瘦素含量低于低LAMA4蛋白组(P<0.05)。结论子痫前期胎盘组织中LAMA4蛋白表达量减少,具体LAMA4蛋白表达水平与滋养细胞侵袭呈正相关,与胎盘缺氧程度呈负相关。 Objective To explore the expression level of laminin subunit alpha 4( LAMA4) protein in placental tissue of preeclampsia patients and its correlation with invasion of trophoblast cells and placental hypoxia. Methods Fifty-six patients with preeclampsia who were treated and gave birth to their babies in the hospital from May 2012 to May 2016 were collceted as observation group; 50 normal pregnant woman receiving prenatal examination and giving birth to their babies in the hospital during the same period were selected as normal control group. According to the expression median of LAMA4 protein in placental tissue,the patients in observation group were further divided into high LAMA4 protein group and low LAMA4 protein group,28 patients in each group. Western Blot was used to detect the expression level of LAMA4 protein in placental tissue; fluorescence quantitative PCR was used to detect the expression level of trophoblast invasion-related gene m RNA in placental tissue; chemiluminescent immunoassay was used to detect the levels of placental hypoxia-related indexes in umbilical cord blood. Results The expression level of LAMA4 protein in observation group was statistically significantly lower than that in normal control group( P〈0. 05); the expression levels of matrix metalloproteinase-9( MMP-9),vascular endothelial growth factor( VEGF),placental growth factor( PLGF) m RNA in high LAMA4 protein group and low LAMA4 protein group were statistically significantly lower than those in normal control group( P〈0. 05); the expression levels of soluble fms-like tyrosine kinase receptor-1( s Flt1) m RNA in high LAMA4 protein group and low LAMA4 protein group were statistically significantly higher than that in normal control group( P〈0. 05); the expression levels of hypoxia-inducible factor-1α( HIF-1α) and HIF proline hydroxylase-1( HPH-1),and leptin in high LAMA4 protein group and low LAMA4 protein group were statistically significantly higher than those in normal control group( P〈0. 05); the expression levels of MMP-9,VEGF,and PLGF m RNA in high LAMA4 protein group were statistically significantly higher than those in low LAMA4 protein group( P〈0. 05); the expression level of s Flt1 m RNA in high LAMA4 protein group was statistically significantly lower than that in low LAMA4 protein group( P〈0. 05); the expression levels of HIF-1α,HPH-1,and leptin in high LAMA4 protein group were statistically significantly lower than those in low LAMA4 protein group( P〈0. 05). Conclusion The expression level of LAMA4 protein in placenta of preeclampsia patients decreases,which is positively correlated with invasion of trophoblast cells and negatively correlated with the degree of placental hypoxia.
