不同药物载体PEDF靶向给药对子宫内膜异位病灶生长及新生血管形成相关因子的影响

摘要 目的在大鼠模型基础上,观察PEDF靶向给药对子宫内膜异位病灶生长及新生血管形成相关因子的影响,并比较不同药物载体的效果。方法采用自体内膜移植法建立子宫内膜异位症大鼠模型,并按计算机随机数字法分为5组,根据干预措施的不同分别为:对照组、无载体组、脂质体组、纳米脂质体组及免疫纳米脂质体组。术后4周打开大鼠腹腔,观察并测量子宫内膜异位病灶的体积,取新鲜病灶组织,采用半定量RT-PCR法检测血管内皮生长因子(VEGF)和凋亡相关因子配体(FAS-L)mRNA的相对表达量。结果无载体组、脂质体组、纳米脂质体组异位病灶体积测量结果与对照组比较差异无统计学意义(P>0.05),而免疫纳米脂质体组的病灶体积均小于以上4组,差异有统计学意义(P<0.05)。对照组、无载体组、脂质体组、纳米脂质体组及免疫纳米脂质体组异位病灶中VEGF mRNA的相对表达量依次降低,各组间差异有统计学意义(P<0.05);对照组、无载体组、脂质体组、纳米脂质体组及免疫纳米脂质体组异位病灶中Fas-L的表达水平依次升高,各组间差异有统计学意义(P<0.05)。结论添加外源性PEDF能够降低子宫内膜异位病灶中VEGF的表达,并促进Fas-L的表达,对新生血管形成及异位病灶生长产生抑制作用。在各种给药载体系统中,以特异性靶向性的免疫纳米脂质体效果最佳。
