摘要 本研究开发软枣猕猴桃EST-SSR引物196对,探究软枣猕猴桃EST-SSR引物在猕猴桃属25个品种中的通用性,并对上述25个品种进行聚类分析。结果表明,筛选得到35对EST-SSR引物,共扩增189条带,平均每对引物扩增5.40条带,条带大小为170~250bp,其中多态性条带达178条(94%),可见软枣猕猴桃EST-SSR引物在猕猴桃属中具有较高的通用性,猕猴桃属植物具有较高的遗传多样性;聚类分析表明,25个猕猴桃属品种的遗传相似系数(GS)为0.591~0.969,在相似系数0.70处,中华猕猴桃与美味猕猴桃归为一支,二者亲缘关系较近。 EST-SSR primers with the number of 196 pairs of Actinidia arguta were developed to screen the transferability of the primers to 25 Actinidia accessions,also cluster analysis of the 25 accessions was constructed. Thirty five pairs of EST-SSR primers were screened out. A total of 189 bands were amplified,with a mean of 5.40bands per primer. The products were 170--250 bp,in which the polymorphic bands were 178 (94%) ,indicating the genetic diversity of Actinidia was high, and the A. arguta EST-SSR primers was highly transferable too. Cluster analysis indicated that the genetic similarity coefficient (GS) of 25 Actinidia accessions ranged from 0. 591 to 0. 969. At the similarity coefficient of 0.70,A. chinensis and A. deliciosa were classified as one branch,showed the close relationship between the two species.
