摘要 目的观察大黄素甲醚对创伤性脑损伤大鼠人脐带间充质干细胞(h UC-MSCs)移植后神经元特异性烯醇化酶(NSE)、胶质纤维酸性蛋白(GFAP)表达的影响。方法将40只2~3个月龄,体重200~250 g清洁级Wistar大鼠随机分为对照组(10只)、颅脑外伤组(10只)、脐带间充质干细胞移植组(10只)、脐带间充质干细胞移植(h UC-MSCs)+大黄素甲醚组(10只)。免疫组织化学法检测神经元特异性烯醇化酶(NSE)、胶质纤维酸性蛋白(GFAP)的表达。结果与模型组比较,h UC-MSCs移植组、联合组大鼠脑组织中神经元特异性烯醇化酶(NSE)、胶质纤维酸性蛋白(GFAP)的表达增强,联合组表达更为显著。结论大黄素甲醚可通过诱导h UC-MSCs表达神经元NSE、GFAP,发挥其对TBI后神经细胞的保护作用。 Objective Effects of physcion on expressions of neuron specific enolase (NSE) and glial fibfillary acidic protein (GFAP) after transplantation of human umbilical cord mesenchymal stem cells (hUC-MSCs) in rats with traumatic brain injury were studied. Methods A total of 40 clean Wistar rats (2 -3 months, weighing 200 - 250 g) were divided randomly into control group ( 10 rats ), TBI group ( 10 rats ), hUC-MSCs group ( 10 rats), and hUC-MSCs transplantation + physeion group ( 10 rats). Expressions of NSE and GFAP were detected by immunohistochemical assay.Results Compared with TBI model group, the expressions of NSE and GFAP in hUC- group and hUC-MSCs transplantation + physcion group were significantly increased, with the hUC-MSCs transplantation + physcion group the most significant. Conclusion Physcion can induce the expression of NSE and GFAP by hUC-MSCs, and exert its protective effect on the neurons after TBI.
