12/15-LOX抑制剂黄芹素对脑缺血/再灌注损伤的保护作用

摘要 目的探讨12/15-LOX抑制剂黄芩素是否具有对抗缺血/再灌注损伤的神经保护作用。方法第一部分,选取健康SD大鼠,线栓法(MCAO)制备脑缺血再灌注/损伤模型,随机分为黄芹素处理组、溶剂对照组(DMSO)及假手术组。分别对各组大鼠进行神经功能评分,采用TTC法测定脑梗死体积,并进行组间比较。第二部分,在大鼠皮层神经元细胞上制备糖氧剥夺(OGD)模型,随机分为OGD+黄芹素处理组、OGD+溶剂对照组(DMSO)及空白对照组。分别采用MTT法和TUNEL法检测各组神经元的细胞活力和凋亡情况,并进行组间比较。结果第一部分的试验结果显示:黄芹素处理组大鼠的神经功能缺损评分低于溶剂对照组(P<0.05),梗死体积百分比也有所降低(P<0.05)。第二部分试验结果显示OGD再合氧后,黄芩素处理组神经元的存活率高于溶剂对照组(P<0.05),同时黄芹素处理组TUNEL阳性细胞比率亦低于对照组(P<0.05)。结论 12/15-LOX抑制剂黄芩素可能具有对抗缺血/再灌注损伤的神经保护作用。 Objective To investigate the possible protective effect of Baicalein, a 12/15-lipoxygenase (12/15-LOX) inhibitor, on the cerebral ischemia and reperfusion (I/R)injury. Methods A transient middle cerebral artery occlusion (MCAO) model was made in SD rats for the first part of the experiment. The animals were divided into three groups: Baicalein treatment group, solvent (DMSO) control group and sham-operation group. The neurological score was assessed and then the infarction volume was measured by 2, 3,5-triphenyltetrazolium chloride (TTC) -staining. In the second part of the experiment, oxygen-glucose deprivation (OGD) model was performed in cultured rat cortical neurons. The cells were divided into three groups: OGD + Baicalein treatment group, OGD + solvent control group and placebo control group. The cell viability and apoptosis was examined by MTT and TUNEL method respectively. Results In contrast to the cases treated with solvent, the neurologic deficits (P 〈 0. 05 ) and infarct volume (P 〈 0. 05 ) was significantly reduced by pretreatment with Baicalein. Meanwhile, the cell viability after OGD in neurons was significantly increased by the treatment with Baicalein (P 〈 0.05) and the amount of TUNEL positive cells in Baicalein treatment group was much lower than that of control group (P 〈 0. 05 ). Conclusion Baicalein, the inhibitor of 12/15-LOX, can improve the neurological deficit and reduce the infarct volume after MCAO in rats. It can also increase the cell viability and inhibit apoptosis after OGD in neurons. Thus Baicalein may have a cerebral protective effect against I/R injury.
