大承气汤对多器官功能障碍综合征大鼠肠源性内毒素血症的疗效机制 被引量:4

Therapy Mechanism of Dachengqi Decoction in Rats with Multiple Organ Dysfunction Caused by Intestinal Endotoxemia

摘要 目的:探讨大承气汤对肠源性内毒素血症的疗效机制。方法:将60只SD大鼠随机分成空白对照组、模型组、大承气汤组、氨苄青霉素组和乳果糖组,每组12只,造模后分别应用生理盐水(空白对照组和模型组)、大承气汤、氨苄青霉素和乳果糖进行灌胃,每12 h 1次,48 h后抽取门静脉血和腹主动脉血,检测内毒素含量、血清TNFa、IL-1、IL-6水平、SOD、LP0水平。结果:(1)大承气汤可明显降低IETM大鼠血中内毒素水平;(2)大承气汤可明显降低TNFa和IL-1水平,大承气汤和乳果糖均可明显降低血清IL-6水平;(3)大承气汤能提高大鼠血清中SOD、LPO水平。结论:大承气汤可通过降低IETM大鼠血中内毒素水平、抑制TNFa、IL-1、IL-6等炎症介质的生成,减轻内毒素对机体的损伤。 Objective: To discuss the therapy mechanism of Dachengqi Decoction in treating intestinal endotoxemia (IETM). Method : 60 SD rats were randomly divided into 5 groups : the blank control group ( group B) , the model group (group M) , Dachengqi Decoction group( group D), Ampicillin group( group A) and lactulose group(group L). MODS models were made by intraperitoneal injection of asepsis Zymosan A. Normal saline, Dachengqi Decoction, Ampicilli- num and lactulose were given to the rats through intragastric administration twice a day after the models were made. Af- ter 48 hours, the endotoxin level of portal vein blood and aortaventrails blood were tested ; TNFa, IL - 1 and IL - 6 in the serum were tested through radioimmunity method, and SOD and LPO were tested through substrate method. Results: 1. Group D can reduce the endotoxin level in IETM rats significantly, compared to group A and group L ( P 〈 0.05 ). 2. The level of TNFa and IL - 1 reduced significantly in group D compared to the other groups ( P 〈 0.05 ). The level of IL -6 reduced significantly in group D and the group L(P 〈0.05). 3. Group D could raise SOD level and reduce LPO level significantly compared to the other groups ( P 〈 0.05 ). Conclusion: Dachengqi Decoction can reduce the body inju- ry caused by endotoxin in IETM rats by reducing the level of TNFa, IL- 1 and IL- 6.
