重组酵母乙肝疫苗免疫蛋鸡后血清抗体与IgY变化规律研究 被引量:1

摘要 研究采用重组酵母乙肝疫苗免疫蛋鸡,探索特异性卵黄免疫球蛋白(IgY)的产生和变化规律。产蛋母鸡经免疫后每周收集所产蛋,分别提取IgY,测定血清抗体效价与抗体的产生量,ELISA定期检测卵黄中的抗体效价变化,Western blotting检测总IgY含量。以未免疫鸡IgY作阴性对照,作血清抗体与IgY变化的动态曲线。取三次免疫后的蛋鸡,解剖分离出各级卵泡,观察各级卵泡发育程度。ELISA结果显示:随着卵泡的生长,卵泡细胞分泌的IgY聚集到卵泡卵黄,卵泡卵黄中IgY的含量在逐渐增加。Western blotting显示:随免疫次数的增加,IgY含量增加。首免后第3天血清抗体被检出,以后逐渐上升,2周后达到高峰,维持到第8周后逐渐下降,到第15周仍可检出抗体。本研究表明:用重组酵母乙肝疫苗免疫蛋鸡,卵黄和鸡血清中可持久保持高效价的特异性抗体,血液中的抗体传递到卵黄需要一定时间,卵黄IgY动态变化规律与血清抗体基本一致。 The production and change of immunogbulines Yolk (IgY)was explored by immunizing recombinant yeast hepatitis B virus (HBV)vaccine in hens. The eggs were collected and extracted IgY after immunization weekly. Serum antibody titers and antibody production were detected. The antibody titer of egg yolk was detected by ELISA, and the total IgY content was detected by Western blotting. The unvaccinated chicken IgY as negative control, the dynamic curve of serum antibody IgY was estabilished. Laying hens was dissected after immunization three times,follicles in the ovaries was isolated, and the degree of development of follicular was observed. The results of ELISA showed that IgY, which was secreted by follicle cells,accumulated into follicular egg yolk and lgY was increased gradually with follicular growth. Western blotting showed that the IgY content was increased with the increase of immunization times. The specific antibodies of total IgY content gradually was increased after immunization. The serum antibodies could be detected at the third day after recombinant yeast HBV vaccine immunization. It reached peak two weeks later and maintained until about the eighth week,and then gradually declined to 15 weeks. After immunization with recombinant yeast HBV vaccine, egg yolk and serum could be sustained to maintain high titers of specific antibodies. It took some time for the antibody in the blood to pass to the yolk. The dynamic changes of yolk IgY were consistent with that of serum antibody.
