牛冠状病毒BCV-Aks-01新疆南疆株全基因组序列测定及其分子特征分析 被引量:7

Sequence and Structural Analysis of the Complete Genome of the Bovine Coronavirus BCV-Aks-01 Strain in Southern Xinjiang,China

摘要 为掌握牛冠状病毒(Bovine coronavirus,BCoV)BCV-Aks-01新疆南疆株的全基因组序列、分子结构特征及遗传变异情况。本研究以新疆南疆某牛场BCV-Aks-01阳性样本为基础,以GenBank中BCoV参考株设计扩增引物和测序引物,提取样本中病毒核酸,对BCV-Aks-01株进行全基因组扩增、测序、构建系统发生树及序列分析。结果表明,BCV-Aks-01新疆南疆株为BCoV,其全基因组长为30 975bp,与GenBank收录的参考株比较,核苷酸同源性高达98.8%,属于β类冠状病毒2a亚类,具有β类冠状病毒共同结构特征。首次获得BCV-Aks-01新疆南疆株全基因组序列并阐明了其基因组分子结构特征。 We wished to study the complete genomic sequence, genomic characteristics, and genetic varia- tion of the bovine coronavirus (BCV)-Aks-01 strain in southern Xinjiang, China. Positive samples were detected by reverse transcription-polymerase chain reaction using special primers. Based on the complete genome of the BCV reference strain, specific primers and sequencing primers were designed, and the com- plete genome of the BCV-Aks-01 strain was amplified and sequenced. Sequence analyses showed that geno- typing of the BCV-Aks-01 strain belonged to betacoronavirus-2a. The 30,975 bp full-length genomic se- quence of the BCV-Aks-01 strain was compiled using DNAStarTM. The sequence of the BCV-Aks-01 strain had 98.8% similarity to the reference strain from GenBank. The BCV-Aks-01 strain had the structural characteristics of betacoronavirus. The BCV-Aks-01 strain is the first BCV reported in southern Xinjiang.
