高果糖摄入对孕鼠淋巴细胞DNA损伤的影响 被引量:1

DAMAGING EFFECT OF HIGH FRUCTOSE INTAKE ON LYMPHOCYTE DNA IN PREGNANT RATS

摘要 目的探讨高果糖摄入对孕鼠淋巴细胞DNA损伤的影响。方法将成年SD雌性与雄性大鼠进行交配,将孕鼠随机分为对照组(CG组)、正常剂量果糖组(NFG组)、高剂量果糖组(HFG组)、高剂量蔗糖组(HSG组)和超高剂量果糖组(SHFG组)5组,各组每天分别给予1 mL去离子水、1.6g/kg果糖水、4.8g/kg果糖水、4.5g/kg蔗糖水和8.0g/kg的果糖水灌胃,连续干预3周,每周称孕鼠体质量,并检测血糖情况。于第3周末腹主动脉取血,利用紫外线分光光度法测定大鼠血清果糖水平,采用碱性单细胞凝胶电泳法(SCGE)测定大鼠外周血淋巴细胞DNA自发损伤和H_2O_2与果糖联合损伤状况。结果与CG组和NFG组比较,HFG组、HSG组和SHFG组孕鼠血清中果糖水平明显增高(F=35.04,P<0.05)。孕3周末,与CG组比较,HFG组、HSG组和SHFG组孕鼠体质量显著增加(F=7.02,P<0.05)。HFG组、HSG组和SHFG组孕鼠的血糖水平明显高于CG组和NFG组(F=6.21,P<0.05)。HFG组、HSG组和SHFG组孕鼠外周血淋巴细胞DNA的自发损伤水平显著高于CG组和NFG组(F=11.62,P<0.05);在5、10和20μmol/L的H_2O_2处理后,HFG组、HSG组和SHFG组孕鼠外周血淋巴细胞DNA损伤水平明显高于CG组与NFG组(F=15.08~75.75,P<0.01)。结论与正常果糖摄入水平相比,孕期过量果糖摄入可引起孕鼠体质量增加、血糖升高;果糖缺乏未见孕鼠外周血淋巴细胞DNA自发及H_2O_2联合损伤增加,而摄入过高剂量果糖和蔗糖时可引发机体外周血淋巴细胞DNA自发损伤及H_2O_2联合损伤加重。 Objective To examine the damaging effect of high-fructose intake on lymphocyte DNA in pregnant rats.Methods Adult female Sprague-Dawleyrats were mated with male rats,and the pregnant rats were then randomly divided into control group(CG),normal-dose fructose group(NFG),high-dose fructose group(HFG),high-dose sucrose group(HSG),and super-high-dose fructose group(SHFG).Pregnant rats in each of the groups received intragastric administration of 1mL deionized water,1.6g/kg fructose water,4.8g/kg fructose water,4.5g/kg sucrose water,and 8.0g/kg sucrose water,respectively.Administration was performed for 3weeks,and body mass and blood glucose of the pregnant rats were measured weekly.Abdominal aorta blood was collected from each animal at the end of week 3,and serum fructose level was measured using UV spectrophotometry.DNA self-damage and H2O2+fructose-induced damage in peripheral blood lymphocytes were assessed using alkaline single cell gel electrophoresis(SCGE). Results The levels of serum fructose in pregnant rats of HFG,HSG,and SHFG were significantly higher than those of CG and NFG(F=35.04,P〈0.05).At the end of week 3,the body mass of pregnant rats in HFG,HSG,and SHFG was significantly higher than that in CG and NFG(F=7.02,P〈0.05),and the levels of fasting blood glucose(FBG)in HFG,HSG and SHFG were also significantly higher than those in CG and NFG(F=6.21,P〈0.05).Peripheral blood lymphocyte DNA self-damage was significantly higher in HFG,HSG and SHFG than in CG and NFG(F=11.62,P〈0.05).Furthermore,peripheral blood lymphocyte DNA damage induced by 5,10 and 20μmol/L H2O2 was also significantly higher in HFG,HSG,and SHFG than in CG and NFG(F=15.08-75.75,P〈0.01). Conclusion High-fructose intake during pregnancy can increase FBG and body mass of pregnant rats.High fructose and sucrose intake,but not fructose deficiency,can exacerbate DNA self-damage and H2O2-induced damage in peripheral blood lymphocytes in pregnant rats.
