摘要
目的:研究不同蔗糖浓度培养基下右旋糖苷酶Dex与氟化钠Na F联用对变异链球菌(S.Mutans)生物膜形成的影响。方法:体外构建S.Mutans生物膜模型。pH电极检测不同蔗糖浓度下处理因素对生物膜培养基p H值的影响;结晶紫染色法检测不同蔗糖浓度下处理因素对早期生物膜形成的影响;扫描电子显微镜观察处理因素对生物膜形成的抑制作用。结果:不同蔗糖浓度下,Dex组对pH值无明显影响(P>0.05),Na F组pH值均大于对照组,差异有统计学意义(P<0.05),Dex+Na F组的p H值大于对照组(P<0.05);除3%和4%浓度组外,其余各浓度组间差异均有统计学意义(P<0.05)。不同蔗糖浓度下,各处理组生物膜形成量均减少,Dex+Na F各浓度组与对照组相比差异均有统计学意义(P<0.05),Dex和Na F对生物膜的形成具有协同抑制作用。在Dex和Na F联用时,生物膜的形成随着蔗糖浓度增加而逐渐减少。结论:Dex和Na F对S.Mutans生物膜的形成具有抑制作用,两者联用具有协同作用,同时蔗糖浓度能够影响生物膜的形成。
Objective:To investigate the effects of exogenous dextranase(Dex) and sodium fluoride (NaF)in different sucrose concentration medium on biofilm formation of Streptococcus mutans ( S. Mutans). Methods: The model of S. Mutans biofilmin in vitro was established. The pH value of the medium with different sucrose concentration during the biofilm forma- tion was detected by pH electrode. Crystal violet staining was used to observe the effects of Dex and NaF on biofilm forma- tion. Scanning electron microscope was applied to detect the inhibitory effect of Dex and NaF on biofilm formation. Results:In different sucrose concentration medium, Dex had no effect on the pH value, but NaF could increase the pH value. The pH val- ues of the Dex + NaF groups were higher than the control groups ( P 〈 0.05 ), and significant difference was existed among Dex + NaF groups with different sucrose concentration( P 〈 O. 05) except the 3% and 4% groups. The mass of biofilm in all treated groups decreased when compared with the control groups, and Dex and NaF had synergistic inhibitory effect on the biofilm formation. There were statistically significant differences in biofilm formation between Dex + NaF groups and control groups( P 〈0.05 ). When simultaneously treated with Dex and NaF ,the formation of biofilm decreased when the sucrose concentration increased. Conclusion: Dex and NaF had inhibitory effect on the biofilm formation of S. Mutans, and they had syn- ergistic effect when used together. Sucrose concentration could also affect the biofilm formation of S. Mutans.
