miR-143在食管鳞癌和上皮内瘤变组织中的表达及其临床意义 被引量:1

摘要 背景:研究发现microRNA在肿瘤的发生、发展过程中起癌基因或抑癌基因的作用。然而,microRNA-143(miR-143)在食管鳞癌中的作用需进一步研究。目的:探讨食管鳞癌组织中miR-143表达及其临床意义。方法:选取2013年1月—2013年12月南京医科大学第一附属医院63例食管鳞癌及其癌旁正常组织以及40例食管上皮内瘤变及其相应正常组织,应用实时定量PCR法检测miR-143表达,并分析其与临床病理特征的相关性。结果:与对照组相比,食管鳞癌和上皮内瘤变组织中miR-143表达均明显下调(P<0.05)。miR-143表达与患者的病理类型有关(P<0.001),而与性别和年龄无关(P>0.05)。miR-143表达与食管鳞癌患者病理分级、淋巴结转移、TNM分期有关(P<0.05),与肿瘤浸润深度无关(P>0.05)。结论:miR-143在食管鳞癌、食管上皮内瘤变组织中的表达均下调,可能与食管鳞癌发生、发展密切相关,有望成为诊断食管鳞癌的新靶点。 Background: Studies have shown that microRNA plays a role of oncogene or tumor suppressor gene in the carcinogenesis and progression of tumor. However,the role of microRNA-143( miR-143) in esophageal squamous cell carcinoma( ESCC) needs further study. Aims: To investigate the expression and clinical significance of miR-143 in ESCC. Methods: Sixty-three ESCC tissues and corresponding para-cancerous tissues,40 esophageal intraepithelial neoplasia( IEN) tissues and corresponding normal tissues from Jan. 2013 to Dec. 2013 at the First Affiliated Hospital with Nanjing Medical University were enrolled. The expression of miR-143 was examined by real-time quantitative PCR,and its correlations with clinicopathological features were analyzed. Results: Compared with controls,expression of miR-143 was down-regulated in ESCC and IEN( P〈0. 05). Expression of miR-143 was correlated with pathological type( P 〈0. 001),but not with gender and age( P〉0. 05). Expression of miR-143 was correlated with pathological staging,lymph node metastasis and TNM staging( P〈0. 05),but not correlated with tumor infiltration depth in ESCC patients( P〈0. 05).Conclusions: Expression of miR-143 is down-regulated in ESCC and IEN tissues,which may be closely related to the development and progression of ESCC,and has the potential to be used as a new target for diagnosis of ESCC.
