混合醇萃取用以降低林可霉素产品中B组分含量的工艺研究 被引量:3

摘要 本文以提高林可霉素产品纯度为目的 ,研究了以混合高碳醇为萃取剂从发酵液中提取林可霉素的工艺过程 ;以混合高碳醇 (80 %辛醇∶2 0 %癸醇 )为萃取剂从发酵液 (pH10 )中提取林可霉素 ,常温下 12级连续逆流操作时 ,萃取率可达 92 .8%;萃取相经过洗涤后 ,以等当量的盐酸反萃 ,反萃液经浓缩、脱色和结晶 ,所得产品中林可霉素B含量 (1.3 %)远低于传统丁醇提取工艺产品 (4 .2 %)。在此基础上 ,还探讨了混合高碳醇萃取和丁醇萃取工艺相结合的可行性 ,以丁醇提取工艺的林可霉素一次结晶为原料 ,通过两级萃取和单级洗涤操作 ,再以盐酸溶液反萃 ,结晶后产品中林可霉素B含量小于 1.8%。 A novel process for extraction of lincomycin from fermentation broth was studied. A mixture of n octanol and n decanol is used as extractant to upgrade the purity of the end product. Extraction yield of 92.8% was obtained in 12 stage counter current extraction of lincomycin from fermentation broth (pH value adjusted to 10) with mixed long chain alcohol (80% n octanol∶20% n decanol). The loaded organic phase was scrubbed and then stoichiometrically stripped with hydrochloric acid. Strip liqour was concentrated, decolorized and mixed with acetone for crystallization of lincomycin. Content of lincomycin B in the final product (1.3%) was much lower than that in product using butanol extraction process (4.2%). Furthermore, the practicability for combination of long chain alcohol and butanol in purification of lincomycin was investigated. Aqueous solution of primary crystallization product in butanol extraction process was extracted in two stage operation and scrubbed in a single stage. The scrubbed organic phase was stripped with hydrochloric acid. Assaying of crystallization product from the strip liquor showed a content of less than 1.8% of lincomycin B with significant quality upgrading.
