摘要
(1)目的探讨微小染色体维持蛋白7(Minichromosome maintenance protein7,MCM7)、S期激酶相关蛋白2(S-phase kinase associated protein 2,SKP2)和细胞周期素依赖性激酶抑制蛋白P27(cyclin dependent kinase inhibited protein,P27)在甲状腺乳头状癌(papillary thyroid carcinoma,PTC)中的表达及其临床病理特征之间的关系。(2)方法采用免疫组织化学S-P法检测MCM7、SKP2和P27蛋白在50例甲状腺乳头状癌、30例结节性甲状腺肿、30例甲状腺腺瘤及20例正常甲状腺组织中的表达。(3)结果甲状腺乳头状癌组织中MCM7、SKP2蛋白的阳性表达率均显著高于结节性甲状腺肿、甲状腺腺瘤及正常甲状腺组织。甲状腺乳头状癌中P27蛋白的阳性表达率显著低于结节性甲状腺肿、甲状腺腺瘤及正常甲状腺组织。MCM7与P27蛋白的表达、SKP2与P27蛋白的表达均呈负相关。(4)结论MCM7、SKP2及P27蛋白与甲状腺乳头状癌的发生和发展起着重要的作用,且MCM7、SKP2蛋白对于淋巴结有无转移方面可能有鉴别意义。在临床上将MCM7、SKP2及P27蛋白联合检测,对于提高术前诊断的准确性及判断淋巴结的转移情况可能有一定的作用。
Objective To explore the expression and significance of MCM7,SKP2 and P27 protein in papillary thyroid carcinoma.Method Detected 50 cases of PTC patients,30 cases of thyroid nodular goiter patients,30 cases of thyroid adenoma patients and 20 cases of normal thyroid patients with immunohistochemical SP method.Results The positive expression rate of MCM7,SKP2 protein in PTC patients were significantly higher than those of thyroid nodular goiter patients, thyroid adenoma patients and normal thyroid patients.The positive expression of P27 protein in papillary thyroid carcinoma patients was significantly lower than those of nodular goiter patients, thyroid adenoma patients and normal thyroid patients.The expression level of P27 protein was negatively correlated with MCM7 and SKP2 protein in papillary thyroid carcinoma patients.Conclusion MCM7,SKP2 and P27 protein play an important role in the development and progression of papillary thyroid carcinoma.MCM7 and SKP2 protein have unique significance in lymph node metastasis. Clinically, the combination of MCM7, SKP2 and P27 protein can improve the accuracy of preoperative diagnosis and determine the lymph node metastasis.
