摘要
目的探讨利用TO型生物制片透明剂替代二甲苯在石蜡组织块脱蜡转透射电镜标本制备的可行性。方法将石蜡组织块分为常规脱蜡组和快速脱蜡组,采用TO型生物制片透明剂代替二甲苯进行脱蜡后作透射电镜标本制备和观察,并与采用二甲苯按照常规脱蜡步骤进行脱蜡后作透射电镜标本制备的结果进行比较。结果 TO型生物制片透明剂常规脱蜡步骤与二甲苯常规脱蜡后转透射电镜标本的切片与观察效果比较,差异无统计学意义(P>0.05)。结论 TO型生物制片透明剂可用于石蜡组织块转透射电镜标本制备时脱蜡,以减少二甲苯对环境和实验者的危害,但其难度略大,标本观察效果略差。
Objective To explore the practicability of T0-type biological transparentizing agent instead of xylene in specimens preparation using paraffin material for transmission electron microscopy. Methods Paraffin-embedded tissue blocks were collected and subdivided into fast dewaxing group and routinely dewaxing group, with the usage of T0-type bjological transparentizing agent instead of xylene in specimen preparation and observation using paraffin material for transmission electron microscopy. A comparison was further performed according to results of routinely xylene dewaxing in specimen preparation using paraffin material for transmission electron microscopy. Results Specimens sections and observed effects showed no significant difference in the compari- son of using T0-type hiological transparentizing agent and xylene in specimen preparation using paraffin material for transmission e- lectron microscopy. In addition, T0-type biological transparentizing agent posed slightly difficulty for slicing after fast dewaxing, and slightly worse observation effect of specimens were also observed subsequently. Conclusion TO-type biological transparentiz- ing agent can be used for specimens" preparation when using paraffin material for transmission electron microscopy to reduce the harm of xylene on the environment and the experimenter,but its difficulty is slightly larger, and the effect of specimen observation is slightly worse.
