摘要
目的探讨转录因子T-bet/GATA-3在早期潜伏梅毒免疫发病机制中的作用。方法应用实时荧光定量PCR检测20例早期潜伏梅毒患者、20例早期显性梅毒患者和25例正常对照者外周血单个核细胞中T-bet和GATA-3 mRNA的相对表达量。结果 (1)潜伏梅毒组T-bet mRNA相对表达量显著低于正常对照组、显性梅毒组,差异有统计学意义(P均<0.05),显性梅毒组与正常对照组比较差异无统计学意义(P>0.05);(2)潜伏梅毒组、显性梅毒组、正常对照组之间GATA-3 mRNA相对表达量差异无统计学意义(P>0.05)。(3)潜伏梅毒组T-bet/GATA-3比值显著低于正常对照组、显性梅毒组,差异有统计学意义(P均<0.05),显性梅毒组与正常对照组比较差异无统计学意义(P>0.05);(4)T-bet、GATA-3mRNA相对表达量与血清RPR滴度之间无相关性(r_s=0.19,r_s=0.10,P均>0.05)。结论 T-bet/GATA-3表达失衡可能在早期潜伏梅毒免疫发病机制中具有重要作用,主要与转录因子T-bet低表达有关。
Objective To investigate the role of transcription factors T -bet/GATA -3 in the immunological pathogenesis of patients with early latent syphilis. Methods The expression levels of T - bet mRNA and GATA - 3 mRNA was determined by real - time quantitative polymerase chain reaction in the peripheral blood from 20 early latent syphilis patients ,20 early symptomatic syphilis patients and 25 healthy controls. Results 1 )The ex- pression level of T- bet mRNA in latent syphilis group was significantly lower than that in symptomatic syphilis group and control group (P 〈 0.05 ) , and there was no difference between symptomatic syphilis group and control group (P 〉 0.05 ). 2) No difference of the expression level of GATA - 3 mRNA was found among latent syphilis group, symptomatic syphilis group and control group ( P 〉 0.05 ). 3 ) The ratio of T - bet/GATA - 3 in latent syph- ilis group was significantly lower than those in symptomatic syphilis group and control group ( P 〈 0.05 ). There was no difference between symptomatic syphilis group and control group ( P 〉 0.05 ). 4) Spearman correlation a- nalysis suggested that there was no association between the expression level of T - bet mRNA or GATA - 3 mRNA and serum RPR titre ( rs = 0.19, P 〉 0.05 ; rs = O. 10, P 〉 0.05 ). Conclusion The imbalance of T - bet/GATA - 3may play an important role in the immunological pathogenesis of early latent syphilis, especially the low expres- sion of T - bet might be involved in the imbalance of T - bet/GATA - 3.
