摘要
试验研究并讨论了3种小麦阿拉伯木聚糖(W9023,W58和W27)分子量及单糖组分的差异。通过高效体积排阻色谱及反向高相液相色谱分别对小麦阿拉伯木聚糖的分子量及单糖组分进行分析。碱提小麦阿拉伯木聚糖重均相对分子量可分为高分子量和低分子量2部分,且分子量大小分别为1.25×10~6~1.49×10~6 g/mol和3.6×10~3~3.8×10~3g/mol。单糖组分分析表明,小麦阿拉伯木聚糖主要有Ara,Xyl,Glu和Gal组成,即表现出典型的AX结构。W9023及W27的Ara/Xyl比值相对较高,分别为0.54和0.51。结果表明:不同来源的小麦阿拉伯木聚糖分子特性有差异。W9023及W27具有相对较高的取代度,更利于后续对阿拉伯木聚糖的研究。
The molecular weight (Mw) and monosaccharide composition of arabinoxylans obtained from three wheat bran sources were studied by high volume exclusion chromatography (HPSEC) and reversed phase high performance liquid chromatography (RP-HPLC), respectively. Each sample contained two populations of AX. The first one consisted of higber Mw AX (1.25× 10^6-1.49× 10^6 g/mol), The second population had a low Mw (3.6× 10^3-3.8× 10^3 g/mol). Monosaccharide composition analysis indicated that wheat bran arabinoxylans were mainly composed of arabinose, xylose, glucose and galactose. The Ara/ Xyl values of W9023 and W27 were 0.54 and 0.51, respectively, which were relatively higher than W58. It could be concluded that the molecular characteristics of three wheat bran ambinoxylans were different according to the reach above. The research provided a reference for the further study of ambinoxylans in the future.
