灯盏花素温敏型鼻用原位凝胶对实验性脑梗死大鼠的药效作用 被引量:8

摘要 目的:探讨灯盏花素温度敏感型鼻用原位凝胶对改善脑缺血的药效作用,为开发灯盏花素鼻用原位凝胶给药系统提供参考依据。方法:将SD大鼠随机分为假手术组、模型组、灯盏细辛静脉注射组、灯盏花素温敏凝胶鼻腔给药高低剂量组、灯盏花素鼻腔给药微球组。在线栓法制备大鼠局灶型脑缺血模型(MCAO)的基础上,进行大鼠神经行为学评分及TTC染色检测大鼠脑梗死面积,评价灯盏花素温度敏感型鼻用原位凝胶对脑缺血保护的药效作用。结果:灯盏花素鼻用原位凝胶高剂量组及其低剂量组的行为学评分与模型组均有显著性差异(P<0.05),与灯盏花素注射液组和微球组比较无显著性差异(P>0.05);鼻腔给药高、低剂量组大鼠的平均脑梗死面积比为7.81%、14.24%,与模型组(20.69%)、静脉注射组(10.81%)和微球组(10.69%)比较均具有统计学意义(P<0.05)。结论:研究表明灯盏花素鼻用温敏凝胶对脑缺血确有改善作用,且优于静脉注射。 Objective:To probe the protective effects of Breviscapine in situ nasa] therrnoreversible gel on the focal cer- ebral ischemia injury in rats in order to provide references evidence of the Breviscapine in situ nasal thermoreversible gel Drug delivery system develop. Methods :Prepare the cerebral artery occlusion(MCAO) model in rats according to the su- ture- occluded method. And then MCAO model SD rats with cerebra/isehernic injury were randomized into the sham - operation group, the model group, Breviscapine solution injection group, Breviscapine in situ nasa/ thermoreversible gel groups( high dose group and low dose group) , Breviscapine nasal administration microspheres group. Measure the neuro- logical deficit score and measure brain infarction area by TTC - staining. Evaluate the pharmaeodynarnic effects of Brevis- capine in situ nasa/thermoreversible gel on the focal cerebra/ischemia injury. Results:There was significant diference be- tween the group of Breviscapine in situ nasal thermoreversible gel and the group of model (P 〈 0. 05 ) in the neuroethology sore, which had no significant diference between the group of hreviscapine solution injection and the group of breviscapine nasal administration microspheres(P 〉0. 05). The area of cerebral infarction percentage of the high Bre content thermo- sensitive gel and the low one was 7.81% and 14.24%. There was significant diference(P 〈0. 05) compared with the group of model(20.69% ) ,the group of breviseapine solution injection( 10.81% ) and the group of breviscapine micro- spheres( 10.69% ). Conclusion:The study proved that Bre thermosensitive gel has definite effect to improve cerebrial is- chemia, better than breviscapine intravenous injection.
