“三穴五针法”对过敏性支气管哮喘大鼠肺组织ERK1/2蛋白表达的影响

Effects of “Three Points and Five Needles” on ERK1/2 Expression in Lung Tissue of Allergic Bronchial Asthmatic Rats

目的:探讨"三穴五针法"对过敏性支气管哮喘大鼠肺组织ERK1/2蛋白表达的影响。方法:将40只SPF级雄性SD大鼠随机分为空白组(C组)、哮喘模型组(M组)、哮喘模型针刺组(A组)及哮喘模型雾化阻断剂组(PD组)4组,每组10只,给予相应干预后,采用HE染色法、免疫组织化学法及Western Blot法观察各组大鼠肺组织病理学变化,检测大鼠肺组织中ERK1/2蛋白的表达程度。结果:A组及PD组大鼠肺组织病理改变较M组均减轻而较C组重,免疫组化法及Western Blot法均显示M组哮喘大鼠肺组织ERK1/2蛋白的表达高于C组(P<0.05),而A组及PD组大鼠肺组织ERK1/2蛋白的表达低于M组(P<0.05)。结论:"三穴五针法"可明显改善过敏性支气管哮喘大鼠炎症反应,抑制哮喘大鼠肺组织ERK1/2蛋白的表达,针刺可能通过抑制ERK1/2蛋白的表达而达到减轻过敏性支气管哮喘症状的目的。

Objective： To investigate the effects of ＂three points and five needles＂ on ERK1/2 expression in the lung tissue of allergic bronchial asthmatic rats. Methods ： Forty SPF grade male rats were randomly divided into control group （C）,asthmatic model group （M）, asthmatic model acupuncture group （A） and asthmatic model spray blocker group （FD）, 10 rats in each group. Groups were given accordingly interventions. HE staining, Immunohistochemistry and West- ern Blot methods were employed to observe the expression of ERK1/2 protein in lung tissue of rats. Results ： HE staining of rat lung tissue indicated that the pathological changes of A group and PD group were slight than those of M group, but were worse than those of C group, hnmunohistochemistry, and Western Blot methods showed that the protein expression of ERK1/2 in lung tissue in M group were higher than that in C group （P 〈0. 05）, but A and PD groups＇ were lower than those of the M group （ P 〈 0. 05 ）. Conclusion： ＂ Three points and five needles＂ can significantly improve the allergic asthmatic rats＇ inflammatory reaction, inhibiting ERK1/2 expression in lung tissue of asthmatic rats, and acupuncture may inhibit the expression of ERKI/2 protein to relieve symptoms of allergic bronchial asthma.
