摘要
目的:探讨调脂一号(TZYH)含药血清对H2O2诱导人脐静脉内皮细胞(HUVECs)炎性因子表达的影响。方法:采用100μmol·L-1H2O2复制HUVECs损伤模型,TZYH含药血清倍比稀释后分为3个剂量干预组(体积分数10%),运用MTT法测定细胞活力,流式细胞仪检测细胞凋亡情况,酶联免疫吸附实验检测细胞分泌白细胞介素1β和肿瘤坏死因子的变化,进一步通过RT-PCR方法检测炎症因子相关基因白细胞介素1β和肿瘤坏死因子的表达。结果:TZYH含药血清能明显提升H2O2损伤后的HUVECs存活率,流式细胞技术结果显示TZYHF含药血清抑制血管内皮细胞凋亡;ELISA和RT-PCR结果提示TZYHF抑制H2O2所致的血管内皮细胞凋亡可能是通过降低IL-1β、TNF-αmRNA和蛋白的表达实现的。结论:TZYH含药血清可以抑制H2O2诱导的内皮细胞凋亡,对血管内皮细胞损伤具有一定的保护作用。
Objective: To investigate Tiaozhi Yihao(TZYH) effect on expression of human umbilical vein endothelial cells (HUVECs) inflammatory factor induced by H2O2. Methods:HUVECs were injuryed by 100 μmol·L^- 1 H2O2, ser- um with TZYH was divided into three dose intervention groups (volume fraction 10% ). MTT method was used to detect the cell activity. The apoptosis was detected by flow cytometry. The expression of interleukin 1 beta and tumor necrosis factor were detected by ELISA and qRT - PCR. Results : Serum with TZYH can be improved survival rate of HUVECs in- jured by H2O2 and Flow cytometry showed that the apoptosis of vascular endothelial cells inhibition by serum with TZY- HF. ELISA and RT- PCR results indicated that TZYHF may inhibit the vascular endothelial cell apoptosis induced by H2O2 by reducing IL - 1β, TNF - α mRNA and protein expression. Conclusion: Serum with TZYH can inhibit apoptosis of endothelial cells induced by H2O2 and has a protective effect on injury of vascular endothelial cell.
