摘要
目的:观察中风胶囊(ZFJN)对大鼠动脉粥样硬化的干预作用。方法:SD大鼠随机分为正常对照组、模型对照组、阿乐对照组以及中风胶囊低、高剂量组。除正常对照组动物摄食正常颗粒饲料,其余各组在摄食高脂饲料的基础上采用注射VD3加激发免疫反应的方法复制AS模型。各组动物连续灌胃给予相应药物90 d,检测各组大鼠血清TC、TG、LDL-C、HDL-C、Ca、SOD、MDA、NO、NOS、ET、IL-18、APN并计算动脉硬化指数(AI)和肝脏指数;取胸主动脉进行HE染色和病理组织学观察。结果:与正常对照组相比,模型组大鼠肝脏指数、AI、血清TC、LDL-C、Ca、MDA、ET、IL-18显著升高(P﹤0.05),而SOD、NO、NOS、APN均有不同程度降低。与模型组比较,ZFJN高剂量组大鼠肝脏指数、AI、血清TC、LDL-C、MDA、ET、IL-18均不同程度降低(P﹤0.05),其血清NO、NOS、APN水平均有显著升高(P﹤0.05)。HE染色显示模型对照组主动脉可见内皮损伤,粥样斑块形成,给药各组均可显著抑制其主动脉损伤。结论:ZFJN通过改善AS模型大鼠脂质代谢、减少氧自由基损伤以及调节血管活性物质等干预AS形成,具有多靶点干预作用。
Objective:To investigate the antiatherogenic effect and possible mechanisms of the Zhongfeng Capsule.Methods:SD rats were randomly divided into 5 groups:normal control group,model control group,Zhongfeng Capsule high and low dose group and positive control group.Normal control group was given saline and basic food,the rest four groups rats were injected VD3 and ovalbumin,while fed with high fat diet.Each group was given the corresponding drugs for 90 days.Serum total cholesterol (TC),triglyceride (TG),low density lipoprotein cholesterol (LDL-C) and high density lipoprotein cholesterol (HDL-C),calcium(Ca),superoxide dismutase (SOD),malondialdehyde (MDA),nitric oxide (NO),nitric oxide synthase (NOS),endothelin (ET),interleukin-18 (IL-18),adiponectin (APN) were determined,visceral index and arteriosclerosis index(AI) were counted,the pathological changes of aorta were observed.Results:The liver index,AI,TC,LDL-C,Ca,MDA,IL-18 and ET were significantly higher while the SOD,NO,NOS,APN levels were lesser in model control group than those in normal control group(P 〈 0.05),compared with those in model group,the liver index,AI,TC,LDL-C,MDA,IL-18 and ET levels were reduced,while the serum NO,NOS and APN levels were raised in the Zhongfeng Capsule high group.HE stain showed aortic endothelial injury and atherosclerotic plaque formation in model group,the positive and Zhongfeng Capsule groups can restrain aortic lesions at different degree.Conclusion:Zhongfeng Capsule can improved metabolism of lipid in model rats,it can reduced oxygen free radical damage and regulated vascular active substances,it had intervention effect on atherosclerotic plaque formation of model animals at multiple targets.
