大黄主要蒽醌类成分的闪式提取及HPLC检测 被引量:2

摘要 目的建立简便的闪式提取法提取中药大黄中蒽醌类成分,并用HPLC法测定其中5种主要游离蒽醌的含量,为大黄蒽醌类化合物高效提取和工业生产提供依据。方法建立闪式提取法提取大黄中蒽醌类成分,与常规的回流提取法进行比较研究,采用C18色谱柱,以乙腈-0.1%磷酸为流动相,梯度洗脱,建立HPLC检测方法,同时检测五种蒽醌的含量,并比较两种方法的提取效率。结果建立的HPLC法能够同时检测5种蒽醌成分,方法准确、可靠。与回流提取方法比较,闪式提取法在短时间内可以达到相近的提取率,显示了闪式提取大黄蒽醌类化合物的优势。结论在相同实验条件下,闪式提取法显示了快速简便、节能高效的特点,可为大黄蒽醌类有效成分成分的高效提取和工业生产提供理论依据。 Objective To establish a simple flash extraction to extract the anthraquinones component of rhubarb and the content of 5 kinds of main free anthraquinones was measured by the HPLC method thus providing basis for the effective extraction and industrial production of anthraquinones component of rhubarb. Methods The anthraquinones component of rhubarb was extracted by the flash extraction, and compared with the routine reflux extraction, and the C18 chromatographic column was used and the acetonitrile-0.1% phosphoric acid was used as the mobile phase and the content of five kinds of anthraquinones was tested by the HPLC test method and the extraction effect was compared. Results Five kinds of anthraquinones could be tested by the HPLC, and the method was accurate and reliable, and the flash extraction could reach the similar extraction rate in a short time, which showed that the advantages of it in extraction of rhubarb anthraquinone analogues. Conclusion The flash extraction is rapid, simple, energy-saving and effective under the same experimental conditions, which can provide theoretical basis for the effective extraction and industrial production of anthraquinones component of rhubarb.
