摘要
目的研究miRNA-193a在肺腺癌患者体内的表达水平,并将其用于肺癌诊断的效能与癌胚抗原(CEA)进行比较,探讨其价值。方法将40例确诊的肺腺癌患者作为肺癌组,同时将40例健康体检者作为对照组。肺腺癌患者中手术12例,术后收集切除的肺段,取正常组织、癌旁组织及癌组织,通过实时荧光定量逆转录PCR(qRT-PCR)检测组织中miRNA-193a的表达水平;检测40例肺癌患者及健康者血清中miRNA-193a和CEA的水平,使用Spearman相关性分析软件分析两者的关系;应用ROC曲线及AUC(95%CI)评估miRNA-193a、CEA作为诊断指标的效能并将两者进行比较。结果肺腺癌患者癌组织miRNA-193a表达量较正常组织明显下降(P<0.05);肺癌组血清miRNA-193a水平较对照组明显下降(P<0.05);而且与CEA之间无相关性(r=0.079,P=0.618),其血清miRNA-193a的AUC为0.829,95%CI为0.741~0.917。结论肺腺癌患者癌组织及血清中miRNA-193a含量均低于正常对照。血清miRNA-193a可能是诊断肺腺癌的一个重要指标。
Objective To explore the diagnostic value of the quantitative detection of serum miRNA-193 ain pulmonary adenocarcinoma.Methods 40 pulmonary adenocarcinoma patients and 40 healthy people were recruited to receive quantitative detection of serum miRNA-193 aand CEA.And then the relationship between the concentration of miRNA-193 aand CEA was analyze by spearman correlation analysis.12 of the 40 patients underwent operation and the different levels of miRNA-193 a expression among pulmonary adenocarcinoma tissues,adjacent tissues and normal tissues was detected by qRT-PCR.The different miRNA-193 alevels were compared among groups of by t-test.The diagnostic performance of miRNA-193 awas evaluated with receiver operating characteristic curves(ROC)and the area under the curve(AUC)and its 95%CI were calculated.Results The relative expressions of miRNA-193 ain pulmonary adenocarcinoma tissues,adjacent tissues and normal tissues were 1.07±0.16;0.80±0.10;0.11±0.07.miRNA-193 aexpression in pulmonary adenocarcinoma tissues was significantly lower than that in other two groups(P<0.05).The expression of serum miRNA-193 ain pulmonary adenocarcinoma group was significantly lower than that in healthy group(P<0.05).There was no correlation between miRNA-193 aand CEA(r=0.079,P=0.618).The detection of miRNA-193 ayielded a ROC AUC of 0.829(0.741-0.917)in discriminating pulmonary adenocarcinoma from healthy group.Conclusion The relative expression of miRNA-193 ain pulmonary adenocarcinoma was significantly lower than that in healthy controls no matter in tissue or serum,serum miRNA-193 amay prove to be a non-invasive biomarker for the auxiliary diagnosis of pulmonary adenocarcinoma.
