肿瘤抑制因子候选基因1在肝癌组织的表达及其临床意义

Clinical significance of tumor suppressor candidate 1 expression in patients with hepatocellular carcinoma

目的探讨肿瘤抑制因子候选基因1（TUSC1）在肝细胞肝癌患者中的表达及其临床意义。方法采用反转录-聚合酶链反应（RT-PCR）及Western blot法检测TUSC1在58例肝细胞肝癌患者中的表达，分析其表达与患者临床生物学特征的关系。结果TUSC1 mRNA表达水平在肝癌中低于相应的癌旁组织表达量（0.31±0.02比0.98±0.03，P=0.002），并且TUSC1 mRNA的表达量与患者的分化程度、肝内转移及门静脉癌栓（PVTT）等病理因素差异有统计学意义（χ^2=17.030、15.730、18.770，P=0.002、0.001、0.001）。而Western blot结果显示，TUSC1在癌组织中蛋白表达量显著低于癌旁组织（120.21±12.35比1 000.31±30.12，P=0.001），其表达水平与患者的病理分化程度、肝内转移和门静脉癌栓明显相关（χ^2=4.613、4.650、6.470，P=0.032、0.031、0.011）。TUSC1 mRNA的表达与其蛋白表达具有明显的相关性（χ^2=10.084, P=0.001）。结论TUSC1在肝癌中表达降低，与患者病理分化程度、肝内转移和门静脉癌栓相关。

Objective To investigate the mRNA and protein expression of tumor suppressor candidate 1, in hepatocellular carcinoma （HCC） and to analyze their clinical significance.Methods Reverse transcription-polymerase and Western blotting technique were used to detect the expression of tumor suppressor candidate 1 （TUSC1） mRNA and protein expression in 58 surgical HCC specimens and matched tumor-adjacent specimens.Results The relative expression level of TUSC1 mRNA was significantly lower in HCC than in tumor-adjacent non-cancerous tissue （0.31±0.02 vs. 0.98±0.03, P=0.002） and was correlated with tumor differentiation, intrahepatic metastasis and portal vein tumor thrombus （PVTT） （χ^2=17.030, 15.730, 18.770, P=0.002, 0.001, 0.001）. TUSC1 protein expression in HCC was significantly lower than tumor-adjacent tissue （120.21±12.35 vs. 1 000.31±30.12, P=0.001） and correlated with tumor differentiation, intrahepatic metastasis and PVTT （χ^2=4.613, 4.650, 6.470, P=0.032, 0.031, 0.011）. There was a significant correlation between the mRNA and protein expression of TUSC1 （χ^2=10.084, P=0.001）.Conclusion TUSC1 expression is downregulated in HCC patients and was associated with tumor differentiation, intrahepatic invasion and PVTT.
