摘要
【目的】揭示牦牛PPARδ基因的遗传变异特征,发现与牦牛生长性状显著相关的候选分子标记,为牦牛分子育种积累基础资料.【方法】以96头牦牛血样为材料,运用DNA池测序和PCR-HRM等技术检测PPARδ基因的序列变异.【结果】检测到牦牛PPARδ基因内含子中2个SNPs.遗传变异特征分析显示,SNP1(g.10045A/G)和SNP2(g.10226A/G)位点属于中度多态性.连锁不平衡和单倍型分析表明,牦牛PPARδ基因2个多态位点之间为弱连锁,单倍型AA属于优势单倍型(频率为42.2%).方差分析表明,单个的SNP与牦牛生长性状有着显著或极显著的关联.【结论】牦牛PPARδ基因的2个SNPs与其生长性状显著相关,此结果可以应用于牦牛的分子育种实践.
【Objective】The research objective was to reveal genetic variation characteristics of bovine PPARδgene and find several candidate molecular markers related to growth traits of yak,thus accumulating some basic information for molecular breeding of yak.【Method】Blood samples collected from 96 yak materials were used to detect the sequence variants within PPARδgene by DNA pool sequencing and PCRHRM technology.【Result】Two novel SNPs within the intron region of PPARδgene were identified in yak.Analysis on genetic variation indicated that SNP1(g.10045A/G)and SNP2(g.10226A/G)loci were moderate polymorphism.Further,the linkage disequilibrium and haplotypes were analyzed between two SNPs,indicating that they were in weak linkage and haplotype of AA was dominant(with frequency of42.2%).The variance analysis showed that the different genotypes of single were all significantly or extremely significantly associated with growth traits of yak.【Conclusion】The two SNPs are significantly correlated to growth traits of yak and the results could apply to molecular breeding of yak.
