人肾脏促红细胞生成素cDNA基因的克隆

摘要 目的:克隆促红细胞生成素(erythropoietin,EPO)c DNA基因,为重组EPO的制备奠定基础。方法:从人肾脏中提取总RNA,采用逆转录-聚合酶链式反应(reverse transcription-polymerase chain reaction,RT-PCR)法扩增EPO c DNA基因后,将其插入到克隆载体p JET 1.2中,DNA测序法鉴定基因序列。结果:对克隆到的EPO c DNA基因进行DNA测序分析鉴定,其结果与Gene Bank中序列完全一致。结论:成功从人肾脏中克隆扩增了EPO c DNA基因,为重组EPO的制备奠定了基础。 Objective:To lay a foundation for the preparation of recombinant erythropoietin (EPO) by cloning EPO cDNA gene. Methods: Total RNA was extracted from human kidney and the reverse transcription - polymerase chain reaction ( RT - PCR) method was adopted to amplify EPO cDNA gene, which was then inserted into a cloning vector pJET 1.2. DNA sequencing method was used to identify gene sequence. Results: The cloned EPO cDNA gene sequence was in accord with sequence in GeneBank through DNA sequencing analysis. Conclusion: EPO cDNA gene from human kidney is successfully cloned and lays a foundation for the preparation of recombinant EPO.
