BrdU标记的BMSCs侧脑室植入后对SAH后CVS大鼠影响的研究 被引量:3

摘要 目的:利用5-溴脱氧尿核苷(bromodeoxyuridine,Brd U)标记的大鼠骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMSCs)植入大鼠侧脑室,观察蛛网膜下隙出血(subarachnoid hemorrhage,SAH)模型大鼠植入BMSCs后由于脑血管痉挛(cerebro vascular spasm,CVS)对大鼠的行为学影响。方法:取原代培养第5代BMSCs,用Brd U标记2 d。采用大鼠自体尾动脉取血枕大池二次注血制作SAH大鼠模型。用Brd U标记好的BMSCs悬液通过大鼠侧脑室注入SAH后的模型大鼠,36只SAH后大鼠随机分为3组,即SAH组(对照组)、SAH后植入DMEM组和SAH后植入BMSCs组,对照组不注射任何物质。术后1 d、2 d、3 d、5 d、7 d检测各组大鼠的神经生物学功能评分,各组取5只大鼠于1 d后进行Morris水迷宫行为学检测。结果:BMSCs组大鼠的生物学功能评分可见在5 d和7 d时,BMSCs组低于其它组,差异有统计学意义(t5d=15.144,P5d=0.042;t7d=12.254,P=0.032)。BMSCs组大鼠平均逃避潜伏期时间要短于SAH组和DMEM组(P<0.05),BMSCs组跨越平台的次数要多于其它两组(t=16.368,P=0.048)。结论:植入BMSCs后能够明显改善SAH后CVS大鼠的感觉运动功能及学习记忆能力。 Objective : To observe the effect of cerebro vascular spasm (CVS) on the behavior of subarachnoid hemorrhage (SAH) model rats transplanted with bromodeoxyuridine(BrdU) labeled bone marrow mesenchymal stem cells(BMSCs) from lateral ventricle injection. Methods: P5 BMSCs was labelled with BrdU for 2 days and SAH model rats were made with twice hemorrhage cistern magna injection method ; SAH model rats were injected with labelled BMSCs cell suspension from lateral ventricles. 36 SAH model rats were randomly divided into the DMEM group, the BMSCs group and the SAH group( the control group). The control group was not injected anything. The neurological scores were detected on 1 d, 2 d, 3 d, 5 d, 7 d. Morris water maze behavior test was performed 1 d later on the 5 rats of every group. Results : The neurological score of BMSCs group was visible on 5d and 7d, which was significantly lower than that of the other two groups( t 5a = 15. 144, P 5a =0. 042 ; t 7a = 12. 254, P = 0. 032 ). Morris mazer BMSCs escaping time is shorter than that of the SAH group and the DMEM group ( P 〈 0.05 ). Times of crossing the plat- form of the BMSCs group were more than those of the other two groups ( t = 16. 368, P = 0. 048 ). Conclusion: After implantation of BMSCs , the sensory motor function and learning and memory ability of CVS rats after SAH could be improved to a certain degree .
