汉坦病毒持续性感染致大鼠乳鼠大脑皮层星形胶质细胞IFN-γ、TNF-α及COX2的表达

摘要 目的:已证实汉坦病毒(hantanvirus,HTV)可持续性感染大鼠乳鼠大脑皮层星形胶质细胞,进一步研究HTV感染致星形胶质细胞IFN-γ、TNF-α及COX2表达变化及意义。方法:汉坦病毒属汉滩病毒(HTNV)76-118和汉城病毒(SEOV)L99分别感染体外培养的大鼠乳鼠大脑皮层星形胶质细胞,用western-blot和RT-PCR检测IFN-γ、TNF-α及COX2表达变化,探究上述因子在HTV持续性感染星形胶质细胞中的意义。结果:通过western-blot和RT-PCR证实HTNV 76-118和SEOV L99株感染离体培养的大鼠乳鼠大脑皮层星形胶质细胞,和未感染病毒组比较,星形胶质细胞IFN-γ和TNF-α表达呈增加趋势(P<0.05),COX2表达无明显变化(P>0.05)。结论:IFN-γ可能参与了HTV所致脑炎的发生或抗病毒作用;TNF-α的表达变化可能与HTV持续性感染大鼠乳鼠大脑皮层星形胶质细胞有关。 Objective :To further study the changes of expressions and significance of IFN-γ、TNF-α and COX2 in astrocytes caused by persistent infection of HTV based on the fact that hantanvirus (HTV) can persistently infect the cortical astrocytes of newborn rats. ncthods:Astro- cytes from cerebral cortex of newborn rats were infected with HTNV 76 - 118 and SEOV L99. Expressions of IFN-γ、TNF-α and COX2 in infec- ted astrocytes were detected by using western - blot and RT - PCR, with the significance of the above factors in HTV persistent infection explored. Results : There is an increasing tendency uf IFN-γ and TNF-α in astrocytes infected by HTNV 76 - 118 and SEOV L99 ( P 〈 0.05 ) ; there was no significant change in the expression of COX2 ( P 〉 0.05 ) . Conclusion: IFN -γ might be involved in encephalitis caused by HTV infection or antiviral effect ; the changes of expressions of TNF - α might be associated with persistently infected cortical astrocytes of newborn rats.
