穴位敷贴治疗与烫伤所致水疱内液体蛋白质组学差异研究

Comparative Proteomics Analysis of Blisters Induced by Acupoint Application Therapy and Scald

目的:运用蛋白质组学分析方法,比较慢性支气管炎患者穴位敷贴过程中产生的水疱与被高温烫伤的患者水疱内液体中的蛋白质组差异。方法:收集15例慢性支气管炎采用穴位敷贴治疗后起疱患者和20例被高温烫伤的患者疱内液体,双向电泳及质谱分析差异蛋白表达。结果:确定穴位敷贴组与烫伤组差异蛋白质点14个,最终成功鉴定13个。其中同一个蛋白对应6个不同的差异蛋白点,有一个差异蛋白点匹配到2个不同的蛋白,因此共得到9个差异表达蛋白,其中5个蛋白表达上调,4个蛋白表达下调。与烫伤组发疱内容物相比,穴位敷贴发疱治疗组的纤维蛋白原β链、纤维蛋白原γ链、半乳凝素-10、触珠蛋白、甲状腺激素结合蛋白的表达较烫伤疱液上调,α1-抗胰蛋白酶、α2-HS糖蛋白、α2巨球蛋白、簇蛋白的表达下调。结论:穴位敷贴治疗与烫伤所致水疱内液体蛋白质成分存在显著差异,差异蛋白主要涉及抗凝血、免疫调节、抗炎、抗凋亡等作用。这提示穴位敷贴发疱可能使机体具有较强的凝血功能、免疫功能、自我修复能力及抗炎抗感染能力,这可能是敷贴发疱治疗呼吸系统疾病的部分物质基础。

Objective：To compare the differences of proteins in blister caused by acupoint application and scald by proteomic analysis. Methods：Liquid of fifteen blisters from patients with chronic bronchitis treated by acupoint application and twenty blisters from scalded patients were harvested,and analysised by the technique of two-dimensional electrophoresis and mass spectrometry analysis. Results：There were fourteen different protein sports between acupoint application group and scalded group. Thirteen differential spots were identified by mass spectrometry and database retrieval. It was suggested that one protein matched to six sports,while two proteins matched to one spot. Thus nine differently expressed protein were identified,among which five were up-regulated while 4 were down-regulated. Compared with scaled group,the protein of fibrinogen beta chain,transthyretin,galectin-10,haptoglobin and fibrinogen gamma chainwere upregulated;α1-antitrypsin,α2-macroglobulin,α2-HS-glycoprotein and clusterin were down-regulated. Conclusion：There were significant differentl proteins in the blister fluid between acupoint application and scald patients,which were related to anticoagulation,immno-regulation,anti inflammation and anti apoptosis. It was indicated that acupoint application might have stronger effects in anticoagulation,immnoregulation,anti inflammation and anti apoptosis. This may be part of material basis of acupoint application treatment for respiratory diseases.
