摘要
[目的]研究姬松茸不同提取物对中波紫外线(ultraviolet B,UVB)致人永生化角质形成细胞(HaCaT)光损伤的保护作用。[方法]采用四甲基偶氮唑蓝(methyl thiazolyl tetrazolium,MTT)法测定不同剂量UVB对正常HaCaT细胞存活率的影响,姬松茸不同提取物对正常HaCaT细胞存活率的影响,以及姬松茸不同提取物对UVB所致光损伤的HaCaT细胞存活率的影响。[结果](1)不同剂量UVB照射后细胞存活率均下降,且呈剂量依赖性,当UVB达20mj/cm2时,细胞存活率为53.23%。(2)姬松茸水提物浓度在0.2~4mg·m L^(-1)、醇提物在0.1~2mg·m L^(-1)、粗多糖在0.2~1mg·m L^(-1)时具有促进HaCaT细胞增殖的作用,醇提物和粗多糖浓度增大则抑制细胞生长。(3)姬松茸水提物浓度在1~4mg·m L^(-1)、醇提物在1~2mg·m L^(-1)、粗多糖在0.5~1mg·m L^(-1)均能显著提高20mj/cm2 UVB照射后HaCaT细胞的存活率(P<0.01)。[结论]姬松茸不同提取物可以在一定程度上减轻UVB诱导的HaCaT细胞的光损伤,具有抗皮肤光老化的潜力。
[Objective] To investigate the protective effects of different extraction of Agaricus blazei Murrill on HaCaT cells damaged by UVB. [Methods] The cell viability which was detected by MTT method was used to determine the impact of different dose of UVB and different extraction of Agaricus blazei Murrill on normal HaCaT cells and HaCaT cells damaged by UVB. [Results](1)The HaCaT cells viability declined after irradiation with UVB, and it was a dose-dependent manner. The cell viability was 53.23% when the dose of UVB was 20mj/cm^2.(2)The water extraction at the concentration of 0.2-4mg·m L^-1,aqueous extraction at the concentration of 0.1-2mg·m L^-1and crude polysaccharide at the concentration of 0.2-1mg·m L^-1could promote the proliferation of HaCaT cells, while aqueous extraction and crude polysaccharide inhibited the proliferation of HaCaT cells at higher concentrations.(3)The water extraction at the concentration of 1-4mg·m L^-1, aqueous extraction at the concentration of 1-2mg·m L^-1and crude polysaccharide at the concentration of 0.5-1mg·m L^-1could enhance the cells viability which was irradiated by 20mj/cm^2UVB(P〈0.01). [Conclusion] The different extraction of Agaricus blazei Murrill all could relieve the photo-damage caused by UVB, and it has the potential of anti-photoaging.
