摘要
目的探讨白藜芦醇(Res)对培养的糖尿病大鼠内皮祖细胞(EPCs)生物学功能的影响。方法采用链脲佐菌素(STZ)腹腔注射联合高糖高脂饮食方法,建立2型糖尿病(T2DM)大鼠模型。造模成功后,Histopaque-1083分离大鼠骨髓单核细胞,种植于EGM-2MV条件培养基中。不同浓度的Res(0,10,25,50μmol/L)孵育第3~5代EPCs 24h,分别采用Edu标记技术、黏附能力测定实验、改良的Boyden小室检测EPCs的增殖、黏附和迁移能力。ELISA法检测EPCs上清液中单核细胞趋化蛋白-1(MCP-1)含量。结果与正常对照组相比,糖尿病大鼠EPCs的增殖、黏附及迁移能力均明显降低,而炎症因子MCP-1的分泌增加;Res可显著提高EPCs的增殖、黏附和迁移能力,同时下调MCP-1的分泌。结论糖尿病大鼠EPCs的生物学功能受损,Res能明显改善糖尿病大鼠EPCs的功能,为临床防治糖尿病心血管并发症提供了理论及实验依据。
Objective To study the effects of resveratrol on the functions of endothelial progenitor cells isolated from diabetic rats. Methods Diabetic rats were induced by intraperitoneal injection of a low dose of streptozotocin( STZ). In the mean time,the rats were fed with high fat and high glucose. Bone marrow mononuclear cells were isolated by Histopaque-1083 and cultured with EGM-2MV. The generations 3th - 5rd of T2 DM EPCs were treated with different concentrations( 0,10,25,50μmol/L) of resveratrol. The proliferation,adhesion,migration and monocyte chemoattractant protein-1( MCP-1) secretion were determined by Edu assay,adhesion test,modified Boyden chamber and ELISA correspondingly. Results EPCs isolated from T2 DM exhibited decreased capabilities of proliferation,adhesion,migration,but had increased release of MCP-1 compared with those from normal rats. Resveratrol promoted the proliferation,adhesion,migration ability and down-regulated the secretion of MCP-1 in a dose dependent manner in T2 DM EPCs. Conclusion The functions of T2 DM EPCs are impaired obviously compared with the normal control. Resveratrol can ameliorate the functions of T2 DM EPCs,and therefore maybe a promise medicine in the prevention or therapy of diabetic vascular disease.
