摘要
目的利用控温烫伤仪建立大鼠烫伤模型和烫伤后感染模型。方法清洁级健康Fisher344大鼠60只,采用随机数字表法分为3组:A组(假烫伤组)10只、B组(烫伤组)20只、C组(烫伤+感染组)30只。A组设定的烫伤条件,压力:1.5kg,温度:37℃,面积:5cm×6cm,时间:15s;B组和C组设定烫伤条件,压力:1.5kg,温度:140℃,面积:5cm×6cm,时间:15s。烫伤24h后,A组和B组在烫伤区域皮下注射生理盐水,C组在烫伤区域皮下注射铜绿假单胞菌。分别于烫伤前1d,注射后12h,24h,3d,7d观察大鼠一般状况、体重、生存状况等变化。在注射后24h和7d每组随机处死6只大鼠,取烫伤皮肤行ELISA检测和石蜡切片、HE染色。结果与A组相比,B组和C组大鼠一般状况、体重、生存率均下降,炎性因子水平上升,皮肤病理切片符合Ⅲ°烫伤表现。与B组相比,C组大鼠一般状况、体重、生存率下降,炎性因子水平上升。结论利用控温烫伤仪成功建立大鼠烫伤模型及烫伤后感染模型,为临床严重烧烫伤患者的治疗提供研究基础。
Objective To establish the scald model and the post-scald infection model by using temperature-controlled scalding instrument. Methods Sixty healthy Fisher 344 rats were randomly divided into three groups: group A( sham group),group B( scald),group C( scald + infection group),group A of 10,group B of 20,group C of 30. Group A was scald,pressure: 1. 5kg,temperature: 37℃,area: 5cm ×6cm,time: 15 s. Group B and Group C were scald,pressure: 1. 5kg,temperature: 140℃,area: 5cm × 6cm,time: 15 s. After 24 hours,group A and group B were subcutaneously injected with normal saline in scalded area,group C was injected subcutaneously with Pseudomonas aeruginosa. The changes of the general condition,body weight and survival status of the rats were observed at 1d before they were scald,and 12 h,24h,3d and 7d after injection. Six rats were sacrificed at 24 h and 7d after injection. The scalded skin was subjected to ELISA and paraffin sections,HE staining. Results Compared with group A,the general condition,weight and survival rate of group B and group C decreased,the inflammatory factor increased,and the skin biopsy accorded with Ⅲ° burn performance. Compared with group B,the general condition,weight,survival rate and inflammatory factors of group C increased. Conclusion The scald model and the post-scald infection model are successfully established by using the scalding scald instrument,which provide the basis for the treatment of severe burn patients.
